苓桂术甘汤通过Thrsp-Srebp1通路改善高脂饮食诱导的大鼠脂肪变性

摘　　要：目的:探讨苓桂术甘汤对高脂饮食诱导的脂肪肝大鼠脂质代谢相关基因表达的影响,阐释苓桂术甘汤改善高脂饮食诱导的脂肪变性的作用机制。方法:24只大鼠随机分成3组,每组8只。正常组予正常饮食喂养8周;模型组高脂饮食饲养8周;苓桂术甘汤组高脂饮食饲养8周,第5周起同时予苓桂术甘汤(3.6g·kg^(-1)·d^(-1))灌胃。8周后,取各组大鼠肝脏组织标本,通过RNA测序筛选差异表达基因,并用RT-PCR及Western Blot对差异基因进行验证。结果:模型组Srebp1、Thrsp、Socs2蛋白水平较正常组显著增加(P<0.01,P<0.05),而苓桂术甘汤可以显著降低Srebp1、Thrsp、Socs2的蛋白水平(P<0.01,P<0.05)。结论:苓桂术甘汤可以通过Thrsp-Srebp1通路改善高脂饮食诱导的脂肪变性,而Socs2也是苓桂术甘汤的作用靶点之一。Objective:To investigate the effects of Linggui Zhugan Decoction(LGZG)on lipid metabolism-related genes,and explain the mechanism of LGZG in improving hepatic steatosis in rats induced by high-fat diet.Methods:Twentyfour rats were randomly divided into 3 group,8 in each one.The normal group was fed with a normal diet for 8 weeks;The model group was fed a high-fat diet for 8 weeks;LGZG group was fed with high-fat diet for 4 weeks,and then given a high-fat diet while LGZG(3.6g·kg^(-1)·d^(-1))was given by intragastric administration for 4 weeks.After 8 weeks,liver tissues were collected,and mRNA was detected by RNA-sequencing.Finally,different expressed genes were verified by RT-PCR and Western Blot.Results:The protein levels of Srebp1,Thrsp and Socs2 in the model group were markedly increased than that in the normal group(P<0.01,P<0.05),while LGZG reduced the protein levels of Srebp1,Thrsp and Socs2 significantly(P<0.01,P<0.05).Conclusion:LGZG could improve the steatosis induced by high-fat diet through the Thrsp-Srebp1 pathway.Socs2 is also one of the targets of LGZG.
