摘  要:目的:考察不同灌流条件下萝卜苷(GRE)的肠吸收特性。方法:采用大鼠在体单向肠灌流模型结合高效液相色谱法,以莱菔子灌流液及G RE灌流液分别灌流,计算低、中、高3种灌流浓度下,G RE在大鼠不同肠段(十二指肠、空肠、回肠、结肠)的表观吸收系数(Papp)。结果:不同实验条件下,GRE的Papp值在(0.75±0.42)~(3.92±0.83)×10^(-3)cm/min之间。采用中浓度GRE灌流液时,吸收最好,各肠段间吸收无显著性差异,Papp值在(3.21±0.42)~(3.92±0.83)×10^(-3)cm/min之间。采用莱菔子灌流液时,GRE最佳吸收肠段是回肠(P<0.05);采用GRE灌流液时,空肠是GRE的主要吸收部位。除回肠段外,其他各肠段在使用GRE灌流液时吸收较好。结论:GRE在肠道中的吸收与灌流浓度和肠段有关,空肠和回肠是主要吸收肠段,纯化后GRE吸收有一定程度提高。Objective:To investigate the absorption characters of glucoraphenin(GRE)in intestinal under different perfusion conditions.Methods:Using high performance liquid chromatography(HPLC),in situ rat single pass intestinal perfusion(SPIP)was carried out to investigate the parameter of apparent absorption coefficient(Papp)in different rat intestinal segments(duodenum,jejunum,ileum,colon)performed with Raphani semen or GRE perfusion under different GRE concentrations(low,middle and high).Results:Under different perfusion conditions,Papp values of GRE were among the range of(0.75±0.42)~(3.92±0.83)×10^(-3)cm/min.Performed with the GRE perfusion of middle concentration,without significant difference in different intestinal segments,the values of Papp were among the range of(3.21±0.42)~(3.92±0.83)×10^(-3)cm/min,which showed the GRE was absorbed best under this condition.Using Raphani semen perfusion,ileum was the main absorption segment(P<0.05);using GRE perfusion,jejunum was the main absorption segment.Except the ileum segment,in the other three absorption segments,GRE showed better absorption characters when performed with GRE perfusion rather than Raphani semen perfusion.Conclusion:The extent of GRE absorption related to the perfusion concentration and the intestinal segment.A better absorption can be achieved when using purified GRE perfusion.
