健脾益气利水汤通过调控OCT-MATE转运系统缓解顺铂所致小鼠肾毒性研究

摘　　要：目的:从OCT-MATE转运系统探讨健脾益气利水汤对小鼠肾损伤模型的疗效及调控机制。方法:通过顺铂诱导的小鼠模肾损伤模型,观察不同给药组健脾益气利水汤对模型体征、肾组织病理和Pt含量以及肾功能的影响,并分析对肾组织内OCT1/2、MATE2分子的基因及蛋白表达。结果:研究表明预防组健脾益气利水汤能提高模型生存率,减少肾脏炎性侵润,显著降低尿液中NAG和KIM-1和血浆中BUN及Cr水平(P<0.05),显著降低Pt在肾脏蓄积(P<0.05),并显著抑制Pt依赖的转运分子OCT-2表达(P<0.05)。结论:健脾益气利水汤通过抑制OCT-2表达从而抑制顺铂在肾脏的进入过程,具有预防降低顺铂肾毒性的作用。Objective:To investigate the therapeutic effect and regulatory mechanism of Jianpi Yiqi Lishui Decoction on renal injury in mice from OCT-MATE transport system.Methods:Through cisplatin-induced kidney injury model of mice,the effects of Jianpi Yiqi Lishui Decoction on the physical signs,renal histopathology,Pt content and renal function were observed,and the gene and protein expression of OCT1/2 and MATE2 molecules in renal tissues were analyzed.Results:The study showed that Jianpi Yiqi Lishui Decoction in the prevention group could improve the survival rate of the model,reduce the inflammatory invasion of the kidney,significantly reduce the levels of NAG and KIM-1 in urine and BUN and Cr in plasma(P<0.05),significantly reduce the accumulation of Pt in the kidney(P<0.05),and significantly inhibit the expression of the Pt-dependent transport molecule OCT-2(P<0.05).Conclusion:Jianpi Yiqi Lishui Decoction can prevent and reduce the nephrotoxicity of cisplatin by inhibiting the expression of OCT-2 and thus inhibiting the entry of cisplatin into the kidney.
