摘 要:目的:研究防纤方对类风湿关节炎肺间质病变(RA-ILD)大鼠的治疗作用并探讨可能机制。方法:Wistar大鼠24只随机分为对照组、模型组、防纤方组;造模后给予相应药物。检测各组大鼠关节炎指数、肿胀程度,观察滑膜及肺组织病理特征,采用实时定量PCR、Western Blot检测Notch信号通路相关蛋白表达及结合情况。结果:与对照组比较,模型组大鼠关节炎指数、肿胀程度明显升高(P<0.01),滑膜及肺组织病理改变明显,肺组织Snail、ZEB-1 mRNA表达水平显著升高(P<0.01),E-Cadherin mRNA表达水平显著下调(P<0.01),Notch1、Jagged1、Jagged2、NICD蛋白表达水平显著上调(P<0.01);而防纤方能改善上述指标变化。结论:防纤方能够有效减轻RA-ILD大鼠关节炎程度,改善肺组织炎性浸润,其作用机制可能与Notch信号通路密切相关。Objective:To study the effects of Fangxian Formula on rheumatoid arthritis-associated interstitial lung disease(RA-ILD)of rats and its mechanism.Methods:A total of 24 Wistar rats were divided randomly into three groups:control group,model group and Fangxian Formula group,then rats were given the corresponding drugs after the modeling.The arthritis index,swelling degree,pathological characteristics of synovium and lung tissue were detected.Using relative quantitative real time PCR,and Western blot analysis,we evaluated protein expressions and binding of Notch signaling pathway.Results:Compared with the control group,the arthritis index and swelling degree in the model group were significantly increased(P<0.01),pathologic changes in the synovium and lung tissues were obvious,while the mRNA expression levels of snail and ZEB-1 were increased(P<0.01)and E-cadherin was significantly decreased(P<0.01),the protein expression of Notch1,Jagged1,Jagged2 and NICD were significantly increased(P<0.01).Fangxian Formula could change the above indexes.Conclusion:Fangxian Formula can reduce the degree of arthritis in rheumatoid arthritis-associated interstitial lung disease effectively of RA-ILD rats,and improve the inflammatory infiltration of lung tissue.Its mechanism may be closely related to Notch signaling pathway.It is considered that Fangxian Formula can change the above indexes.
