瑶医水莲风亏方对盆腔炎性疾病后遗症大鼠IL-6、IL-8、VEGF及MMP-2表达的影响

摘　　要：目的:探讨瑶医水莲风亏方对盆腔炎性疾病后遗症(SPID)大鼠白细胞介素(IL)-6、IL-8、血管内皮生长因子(VEGF)及基质金属蛋白酶-2(MMP-2)表达的影响及其可能机制。方法:将50只造模后的SD大鼠随机分为模型组、妇科千金片组和瑶药高、中、低剂量组,另设空白组10只。空白组、模型组给予0.9%氯化钠溶液灌胃,其他治疗组以相应药物灌胃治疗。21d后使用HE染色观察子宫组织形态学改变;采用酶联免疫吸附(ELISA)法检测血清中细胞因子IL-6、IL-8的含量;Western Blot检测各组大鼠子宫组织内VEGF、MMP-2蛋白表达水平。结果:与空白组比较,模型组大鼠血清中IL-6、IL-8含量显著升高(P<0.05),大鼠子宫组织中VEGF和MMP-2蛋白表达显著升高(P<0.05);与模型组比较,各给药组大鼠血清IL-6、IL-8含量显著降低(P<0.05),大鼠子宫组织中VEGF和MMP-2蛋白表达显著降低(P<0.05)。结论:水莲风亏方可能通过下调大鼠血清中炎症因子及抑制相关蛋白的表达,减少盆腔内炎症反应,可有效治疗SPID。Objective:To investigate the effect of Yao medicine Shuilian Fengkui Formula on the expression of IL-6,IL-8,VEGF and MMP-2 in rats with sequelae of pelvic inflammatory disease(SPID)and its possible mechanism.Methods:Fifty SD rats were randomly divided into model group,Fuke Qianjin Tablet group,high-,medium-,low-dose Yao medicine group.The blank group was given equal volume distilled water by intragastric administration,the model group was given normal saline intragastric administration,and the other treatment group was treated with corresponding drugs.HE staining was used to observe the histological changes of uterus after 21 days.The contents of cytokines IL-6 and IL-8 in serum were determined by ELISA assay.The expression levels of vascular endothelial growth factor VEGF and MMP-2 proteins in the uterine tissues of rats in each group were detected by Western Blot.Results:Compared with the blank group,the serum levels of IL-6 and IL-8 in the model group were significantly increased(P<0.05),and the expressions of VEGF and MMP-2 in the rat uterus were significantly increased(P<0.05).Compared with the model group,all giving drug groups significantly reduced the contents of IL-6 and IL-8 in the serum of rats(P<0.05),and significantly inhibited the expression of VEGF and MMP-2 in the uterine tissues of rats(P<0.05).Conclusion:Shuilian Fengkui Formula may reduce the inflammatory reaction in the pelvic cavity by down-regulating the expression of inflammatory factors and inhibiting related proteins in the rat serum,and can effectively treat SPID.
