摘  要:目的:调查海淀区新型冠状病毒(以下简称"新冠病毒")核酸检测实验室污染情况,并规范出现污染后的处置措施。方法:对海淀区34家新冠病毒核酸检测实验室内物表进行涂抹采样,运用real time RT-PCR方法进行新冠病毒核酸检测。结果:海淀区34家新冠病毒核酸检测实验室采集的510件样本中,有1件新冠病毒核酸检测结果为阳性,推测污染原因为八连管管盖崩开导致核酸扩增仪内部被阳性对照扩增后的基因片段污染。发现污染后对该实验室采取了检测结果追溯、清洁消毒、设备耗材更换、人员培训、消毒效果验证及盲样考核验收等措施,可以有效去除实验室内核酸污染。结论:海淀区新冠病毒核酸检测实验室生物安全风险总体可控,通过人员培训、制度撰写、消毒清洁效果验证等措施可以提高检测机构处置实验室污染的能力。建议所有实验室加强生物安全管理,并定期开展实验室污染监测。Objective To assess the contamination status in nucleic acid testing laboratories for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in Haidian district and to standardize the disposal measures after contamination.Methods Surface swab specimens of subjects inside 34 nucleic acid testing laboratories for SARS-CoV-2 in Haidian district were collected.Real time RT-PCR method was used to detect nucleic acids of SARS-CoV-2.Results A total of 510 specimens were collected.One sample was tested positive for nucleic acid of SARS-COV-2.The possible reason could be the inner surface of the PCR instrument being contaminated by the amplicon of positive control because the caps of PCR strip tubes were opened.After the contamination was found,tracing of test results,decontamination,replacement of equipments and consumables,personnel training,disinfection effect verification and blind sample assessment were used to effectively remove nucleic acid contamination in the laboratory.Conclusions The biosafety risk of nucleic acid testing laboratories for SARS-CoV-2 in Haidian district was generally controllable.Measures such as personnel training,documentation and disinfection and cleaning effect verification can improve the ability of testing institutions to deal with laboratory contamination.It is recommended that all laboratories strengthen biosafety management and regularly carry out laboratory contamination monitoring.
