摘 要:目的:观察温经止痛合剂对子宫内膜异位症(EMs)模型大鼠活丝裂原活化蛋白激酶(MAPK)通路相关因子细胞外信号调节激酶1(ERK1)和c-Jun氨基末端激酶(JNK)表达的影响。方法:将90只雌性成熟未交配过的Wistar大鼠随机分为6组,即正常组,模型组,西药组,中药低、中、高剂量组,各15只。异位造模成功后分别给予高、中、低剂量温经止痛合剂、西药(孕三烯酮胶囊)及0.9%氯化钠溶液灌胃,正常组予0.9%氯化钠溶液灌胃。RT-PCR法及Western Blot法检测大鼠在位、异位内膜中ERK1、JNK的mRNA和蛋白表达。结果:模型组大鼠在位内膜上ERK1、JNK的mRNA及蛋白表达显著高于正常组(P<0.01);中药高、中剂量组和西药组在位、异位内膜上ERK1、JNK的mRNA和蛋白表达显著低于模型组(P<0.01)。结论:EMs的发生、发展与MAPK信号通路有关,温经止痛合剂可抑制MAPK信号通路相关因子ERK1、JNK的表达,抑制异位内膜的黏附、侵袭及血管生成。Objective: To observe the effects of Wenjing Zhitong Mixture on the expression of extracellular signalregulated kinase 1(ERK1) and c-Jun amino-terminal kinase(JNK) related to mitogen-activated protein kinase(MAPK) pathway in endometriosis(EMs) model rats. Methods: Ninety mature female unmated Wistar rats were randomly divided into 6 groups, namely normal group, model group, western medicine group, low, medium and high dose groups of Chinese medicine, each with 15 rats. After successful heterotopic modeling, high, medium and low doses of Wenjing Zhitong Mixture, western medicine(gestrinone capsule) and 0.9% sodium chloride solution were given by gavage, while the normal group was given 0.9% sodium chloride solution by gavage. RT-PCR and Western Blot were used to detect the mRNA and protein expression of ERK1 and JNK in eutopic and ectopic endometrium of rats. Results: The mRNA and protein expressions of ERK1 and JNK in the model group were significantly higher than those in the normal group(P<0.01). The mRNA and protein expressions of ERK1 and JNK in eutopic and ectopic endometrium in high and medium dose groups of Chinese medicine and western medicine group were significantly lower than those in model group(P<0.01). Conclusion: The occurrence and development of EMs is related to MAPK signaling pathway. Wenjing Zhitong Mixture can inhibit the expression of ERK1 and JNK related to MAPK signaling pathway, and inhibit the adhesion, invasion and blood vessels of ectopic endometrium Generate.
