摘  要:目的:研究逍遥散合八珍汤对自发乳腺癌小鼠肿瘤生长的影响及分子机制。方法:自发乳腺癌模型随机分成模型组,紫杉醇组,逍遥散合八珍汤低、中、高剂量组各10只,给药直至濒死期;另设正常组(未发瘤)10只,灌胃0.9%氯化钠溶液;采用苏木素-伊红(HE)染色法观察各组肿瘤组织形态,Western Blot法分析肿瘤组织中丝裂原激活蛋白激酶激酶激酶1(MAP3K1)、叉头框家族D3基因(FOXD3)、Krüppel样因子4(KLF4)、性别决定区Y框蛋白2(SOX2)的蛋白表达。结果:与模型组比较,紫杉醇组和逍遥散合八珍汤中、高剂量组肿瘤小鼠的生存期显著延长(P<0.05),肿瘤组织重量显著下降(P<0.01)、组织形态改善;与模型组比较,紫杉醇组和逍遥散合八珍汤中、高剂量组MAP3K1、FOXD3、KLF4的蛋白表达显著上升,SOX2蛋白表达显著降低(P<0.05)。结论:逍遥散合八珍汤可通过上调MAP3K1、FOXD3、KLF4蛋白表达,下调SOX2蛋白表达来调控ERK/MAPK信号通路,从而抑制肿瘤的生长。Objective: To study the effects and molecular mechanism of Xiaoyao Powder combined with Bazhen Decoction on tumor growth in spontaneous breast cancer mice. Methods: Spontaneous breast cancer mouse models were randomly divided into model group, paclitaxel group, Xiaoyao Powder combined with Bazhen Decoction low, medium and high dose groups of 10 mice each, and they were administered until the dying period;another 10 normal group(no tumor) mice gavage 0.9% sodium chloride solution. Hematoxylin-eosin(HE) staining was used to observe the tumor morphology of each group. Western Blot was used to analyze the protein expression of mitogen-activated protein kinase kinase kinase kinase 1(MAP3 K1), forkhead box family D3 protein(FOXD3), Krüppel-like factor4(KLF4), sex-determining region Y-box protein 2(SOX2) in tumor tissues. Results: Compared with the model group, the survival time of tumor mice in the paclitaxel group, Xiaoyao Powder combined with Bazhen Decoction medium and high dose groups was significantly prolonged(P<0.05), the tumor tissue weight was significantly reduced(P<0.01), and the tissue morphology was improved. Compared with the model group, the protein expressions of MAP3 K1, FOXD3, and KLF4 in the paclitaxel group, the medium-dose, and high-dose of Xiaoyao Powder combined with Bazhen Decoction group significantly increased, and the expression of SOX2 protein decreased significantly(P<0.05). Conclusion: Xiaoyao Powder combined with Bazhen Decoction can regulate the ERK/MAPK signal pathway by up-regulating the expression of MAP3 K1, FOXD3, KLF4 protein and down-regulating the expression of SOX2 protein, thereby inhibiting tumor growth.
