摘 要:目的:研究白术提取物对马钱子中马钱子碱和士的宁药代动力学的影响。方法:采用LC-MS检测大鼠单次给药后不同时间点的血药浓度,DAS 3.0软件计算药代动力学参数,SPSS Statistics 21分析配伍白术前后马钱子生物碱主要药代动力学参数的差异。结果:配伍白术后,马钱子2种生物碱在大鼠血浆中的主要药代动力学参数AUC0-t、AUC0-∞均显著增加(P<0.05,P<0.01),表观分布容积V_(z)/F、清除率CL_(z)/F显著减少(P<0.05,P<0.01),马钱子碱Cmax显著增加(P<0.05)。结论:白术可以抑制马钱子生物碱的分布,减少马钱子生物碱的消除,提高马钱子生物碱的血药浓度。Objective: This study aimed to investigate the influence of Baizhu extract on the pharmacokinetics of two strychnine alkaloids, i.e. brucine(BRU) and strychnine(STR) from Maqianzi. Methods: The rats plasma concentration of the drugs were determined by LC-MS for dose response and time dependent curves. DAS 3.0 was used to calculate the pharmacokinetic parameters, and the differences of main pharmacokinetic parameters between the two groups were analyzed by SPSS Statistics 21. Results: Comparing with Maqianzi group, the AUC0-t, AUC0-∞, and Cmax of two alkaloids in Maqianzi-Baizhu group was significantly increased(P<0.05, P<0.01), and the Vz/F and the CLz/F was decreased(P<0.05, P<0.01). Conclusion: Baizhu can inhibit the distribution of strychnine alkaloids to the whole body, increase the plasma concentration of strychnine alkaloids in the body, and reduce the elimination of strychnine alkaloids in the body.
