摘  要:目的:探究益气活血法代表方暖心康通过线粒体能量代谢途径改善缺血再灌注致慢性心力衰竭小鼠心肌纤维化的作用与机制。方法:随机将30只SPF级C57BL/6J雄性小鼠分为假手术组、模型组与暖心康组,采用冠状动脉结扎的方法制备小鼠心肌缺血-再灌注损伤模型(I/R)。Masson染色检测各组小鼠心脏胶原纤维沉积情况,透射电镜观察小鼠心脏线粒体结构,JC-1流式检测线粒体膜电位水平,PCR检测PGC-1α、CPT-1、GLUT4、MPC1 mRNA表达水平的变化。结果:与假手术组比较,模型组小鼠心脏形态增大,胶原纤维沉积明显增多;线粒体超微结构遭到破坏,心肌细胞线粒体膜电位下降,CPT-1 mRNA水平显著升高(P<0.05),PGC-1α、GLUT4、MPC1 mRNA表达水平显著降低(P<0.05,P<0.01);与模型组比较,暖心康组减轻心衰小鼠心脏胶原沉积,保护线粒体结构,提高心肌细胞线粒体膜电位,CPT-1 mRNA水平显著下降(P<0.01),PGC-1α、GLUT4、MPC1mRNA表达显著升高(P<0.01)。结论:益气活血法代表方暖心康减轻缺血再灌注致慢性心力衰竭小鼠心肌纤维化,可能是通过改善线粒体能量途径实现的。Objective: To explore the role and mechanism of Nuan xinkang, a representative of the method of replenishing qi, promoting blood circulation, in improving myocardial fibrosis in mice with chronic heart failure induced by ischemia-reperfusion through mitochondrial energy metabolism. Methods: Thirty SPF C57 BL/6 J male mice were randomly divided into sham operation group, model group, Nuanxinkang group, prepared by coronary artery ligation mouse myocardial ischemia-reperfusion injury model(I/R). Masson staining to detect the deposition of collagen fibers in the hearts of mice in each group, transmission electron microscopy to observe the structure of mice heart mitochondria, JC-1 flow cytometry to detect the level of mitochondrial membrane potential, and PCR to detect the expression levels of PGC-1α, CPT-1, GLUT4 and MPC1 mRNA variety. Results: Compared with the sham operation group, the mice heart shape in the model group increased, and collagen fiber deposition increased significantly. The ultrastructure of mitochondria was destroyed, the mitochondrial membrane potential of cardiomyocytes decreased, the level of CPT-1 mRNA increased(P<0.05), and the expression levels of PGC-1α, GLUT4, MPC1 mRNA decreased(P<0.05, P<0.01);Compared with the model group, Nuanxinkang group reduced cardiac collagen deposition in mice with heart failure, protected mitochondrial structure, increased myocardial cell mitochondrial membrane potential, and significantly decreased CPT-1 mRNA levels(P<0.01). The expression of PGC-1α, GLUT4, MPC1 mRNA was significantly increased(P<0.01). Conclusion: Nuanxinkang, the representative of the method of replenishing qi, activating blood can reduce myocardial fibrosis in mice with heart failure caused by ischemia reperfusion. The mechanism may be achieved by regulating mitochondrial energy pathways.
