摘 要:目的:探讨精制保和颗粒(RBG)对大肠癌作用的分子机制。方法:采用网络药理学技术分析RBG治疗大肠癌关键成分及靶标,并通过体外实验进一步研究RBG对大肠癌细胞HCT-116和HCT-8生长抑制的作用机制。结果:利用相关数据库及拓扑分析共筛选出22个潜在活性成分和217个潜在靶点,靶点富集分析发现与大肠癌等多种肿瘤和多条肿瘤相关通路有关。体外研究发现,RBG能明显抑制HCT-116和HCT-8细胞活力、集落形成数量和诱导细胞凋亡,并能显著下调Bcl-2和上调Bax蛋白表达,促进Caspase-3和Caspase-9活化,同时显著抑制AKT信号通路的活化。结论:RBG具有多成分、多靶点和多途径的作用特点,可通过诱导细胞凋亡抑制大肠癌细胞的生长,且抑制AKT信号通路的活化可能是其重要的作用机制。Objective:To investigate the molecular mechanisms of Refined Baohe Granules(RBG)in the treatment of colorectal cancer(CRC).Methods:Using network pharmacology technology to analyze the key components and targets of RBG in the treatment of CRC,and further study the mechanism of RBG on the growth inhibition of CRC cells through in vitro experiments.Results:Based on the network pharmacology analysis and network topology analysis,22 potential active compounds and 217 predicted targets were obtained.Enrichment analysis revealed that they were related to multiple tumor and tumor-related pathways,such as CRC.In vitro study showed that RBG significantly inhibited the cell viability and the number of colonies formation,as well as induced apoptosis in HCT-116 and HCT-8 cells.Moreover,RBG treatment remarkably down-regulated Bcl-2 expression and up-regulated Bax expression,promoted the activation of Caspase-3 and Caspase-9,while inhibiting the activation of the AKT signaling pathway.Conclusion:RBG exhibits a potential on anti-CRC with the characteristics of multiple compounds,multiple targets and multiple signaling pathways.RBG can inhibit the growth of CRC cells by inhibiting cell proliferation and promoting cell apoptosis through suppressing the activation of AKT signaling pathway.
