摘 要:目的:探讨三七重楼生化汤(SCSD)促进大鼠子宫切口早期愈合机制。方法:30只大鼠手术建模,随机分为对照组(0.9%氯化钠溶液)、SCSD-L组(SCSD 30g·kg-1·d-1)和SCSD-H组(SCSD 60g·kg-1·d-1),灌胃10d,收集血清,取子宫切口标本。HE染色观测局部炎症反应及肌层厚度,Masson染色观察肌纤维排列。小鼠成纤维细胞予SCSD水提物及含药学清干预24h,Western Blot、免疫荧光及RT-qPCR检测血管内皮生长因子(VEGF)、转化生长因子β(TGF-β)、CollagenⅢ蛋白/mRNA表达。结果:(1)SCSD减轻子宫切口炎症反应,增加肌层厚度,使肌纤维排列有序。(2)浓度为2.5、5、10mg/mL的SCSD可上调VEGF蛋白表达(P<0.01);浓度为10mg/mL的SCSD可上调CollagenⅢ蛋白表达(P<0.05);浓度为2.5、5mg/mL的SCSD可上调VEGF mRNA表达(P<0.05,P<0.01),且浓度越高升高越显著(P<0.05);浓度为5mg/mL的SCSD可上调CollagenⅢmRNA表达(P<0.01);不同浓度SCSD水提物对TGF-β蛋白/mRNA表达未见显著性影响。(3)SCSD-L、SCSD-H含药血清均可上调VEGF、CollagenⅢ蛋白表达(P<0.05),VEGF、TGF-βmRNA表达(P<0.05,P<0.01);SCSD-H含药血清可上调CollagenⅢmRNA表达(P<0.05)。结论:SCSD通过改善子宫切口局部病理变化及VEGF、TGF-β、CollagenⅢ表达,从而促进子宫切口早期愈合。Objective: To observe the mechanism of Sanqi Chonglou Shenghua Decoction(SCSD) in promoting the early healing of uterine incision. Methods: Thirty rats were operated and randomly divided into control group(normal saline), low dose group(SCSD-L, 30 g·kg-1·d-1) and high dose group(SCSD-H, 60 g·kg-1·d-1). After 10 days of intragastric administration, the blood and the uterine incision samples were collected. The inflammatory reaction and myometrium thickness of uterine incision were observed by HE staining, the arrangement of uterine muscle fibers was observed by Masson staining. Mouse fibroblasts were treated with SCSD for 24 hours. Western Blot, immunofluorescence and RT-qPCR were used to detect the expression of VEGF, TGF-βand Collagen Ⅲ protein/mRNA. Results:(1)SCSD reduced inflammation, increased the thickness of myometrium, and made the muscle fibers arranged more orderly.(2)The expression of VEGF protein increased significantly when the concentration of SCSD was 2.5, 5, 10 mg/mL(P<0.01);The expression of CollagenⅢ protein increased significantly when the concentration of SCSD was 10 mg/m L(P<0.05);The expression of VEGF mRNA increased significantly when the concentration of SCSD was 2.5, 5 mg/mL(P<0.05, P<0.01), and the increase was more significant in a higher concentration(P<0.05);The expression of CollagenⅢ mRNA increased significantly when the concentration of SCSD was 5 mg/mL(P<0.01);different concentrations of SCSD have no significant effect on the expression of TGF-β protein/mRNA.(3)The expression of VEGF, CollagenⅢ protein and the expression of VEGF, TGF-β mRNA increased significantly in SCSD-L, SCSD-H containing serum groups(P<0.05, P<0.01);The expression of CollagenⅢ m RNA increased significantly in SCSD-H containing serum group(P<0.05). Conclusion: SCD promoted the early healing of uterine incision by improving the pathological changes of uterine incision, increasing the expression of VEGF, TGF-β and Collagen Ⅲ.
