摘 要:目的:运用基于同位素标记相对和绝对定量技术(iTRAQ)的蛋白质组学分析玄参水煎液干预阴虚火旺甲状腺功能亢进(简称"甲亢")的滋阴降火作用。方法:将SD大鼠分为空白组、左甲状腺素钠片(优甲乐)诱导的阴虚火旺甲亢模型组(模型组)及玄参组。基于iTRAQ技术的蛋白质组学分析用于探讨各组中蛋白质表达水平的结果。结果:通过iTRAQ分析,鉴别出了3 981种蛋白质,其中142种在模型组中发生差异表达并被认为是潜在生物标志物。玄参干预后,玄参表现出了纠正78种异常表达蛋白质的趋势,涉及碳代谢、氨基酸的生物合成、淀粉和蔗糖代谢等代谢途径,与ErbB信号通路相关。结论:玄参对优甲乐诱导的阴虚火旺甲亢大鼠中蛋白质通路具有调节作用,其可能成为治疗或预防阴虚火旺甲亢的候选药物。Objective:To investigate the effects of Radix Scrophulariae(RS)on hyperthyroidism rat model of yin deficiency with effulgent fire by proteomics analysis based on iTRAQ technology.Methods:SD rats were divided into control,hyperthyroidism rat model of yin deficiency with effulgent fire,and hyperthyroidism+RS groups.iTRAQ-based proteomics based on proteomics research approach was used to investigate the changes in the expression levels of proteins in different groups.Results:Through the iTRAQ analysis,3981 modulated proteins were identified,out of which 142 were found to be altered in hyperthyroidism rat model group and considered as potential biomarkers.After treatment with RS,the hyperthyroidism+RS groups showed the tendency to correct the abnormal expressions of 78 proteins out of 142 potential biomarkers,which were associated with the formation of carbon metabolism,amino acid biosynthesis,starch and sucrose metabolism,ErbB signaling pathway,etc.Conclusion:Radix Scrophulariae may be a promising candidate for prevention or treatment of hyperthyroidism by regulating the related proteomic pathways in hyperthyroidism rat model of yin deficiency with effulgent fire.
