摘 要:目的:基于网络药理学探讨青光安Ⅱ号方治疗青光眼的分子机制。方法:通过TCMSP筛选和预测青光安Ⅱ号方潜在的入血活性成分和作用靶点;通过疾病基因数据库挖掘青光眼相关的靶点;采用STRING数据库绘制成分-靶点和疾病-靶点的PPI网络,并提取这两个网络的交集;采用DAVID 6.8数据库分析筛选关键靶点与作用机制。结果:在TCMSP数据库共检索出与青光安Ⅱ号方相关的潜在活性成分808个,根据药代动力学参数筛选获得102个潜在入血活性成分,并检索出与这些成分相关的靶点1 018个;从疾病基因数据库检索并筛选得到与青光眼直接相关的靶点82个;成分靶点和疾病靶点PPI网络取交集后得到以乙酰胆碱受体、肾上腺素受体为主的14个关键蛋白;这些蛋白主要涉及到神经活性配体-受体相互作用、钙信号通路等。结论:青光安Ⅱ号方能通过乙酰胆碱受体、肾上腺素受体等14个关键蛋白治疗青光眼,其机制可能与降低眼压相关。Objective: To explore the molecular mechanism of Qingguangan Ⅱ Formula for glaucoma based on network pharmacology. Methods: Screening and prediction of possible blood-supplying active ingredients and gene targets of Qingguangan Ⅱ Formula through TCMSP;mining gene targets related to glaucoma through the disease gene database;using STRING database to draw component-target and disease-target PPI Network, and extract the intersection of these two networks;use DAVID database analysis to screen key targets and action mechanism. Results: A total of 808 active ingredients related to Qingguangan Ⅱ Formula were retrieved from the TCMSP database. A total of 102 blood-supplying active ingredients were obtained by screening based on pharmacokinetic parameters, and 1018 targets related to these ingredients were retrieved. From the disease Gene database search for disease screening revealed 82 genes directly related to glaucoma;14 key proteins were obtained after the intersection of the component target and disease target PPI network;These proteins are mainly involved in neuroactive ligand-receptor interaction, calcium signaling pathway, etc. Conclusion: Qingguangan Ⅱ Formula can treat glaucoma through 14 key proteins, such as acetylcholine receptor and adrenergic receptor, and its mechanism may be related to lowering intraocular pressure.
