摘 要:目的:探究利拉鲁肽对脂多糖(LPS)诱导大鼠肺微血管内皮细胞PI3K/Akt信号通路的影响。方法:使用LPS诱导培养成功的大鼠肺微血管内皮细胞为实验对象,将其差速贴壁实施纯化,使用不同浓度的利拉鲁肽(0、1、10、100、1000 nmol/L)对细胞进行干预,MTT法绘制细胞生长曲线,分析不同浓度利拉鲁肽对细胞生长影响;采用Western blot法分别检测正常细胞组、利拉鲁肽(100 nmol/L)及PI3K/Akt通路阻断剂(LY294002)对p-Akt、Akt蛋白表达的影响;采用Brdu荧光标记法及划痕实验检测利拉鲁肽对细胞增殖和迁移能力的影响。结果:①通过绘制细胞生长曲线发现,100 nmol/L利拉鲁肽为细胞最适生长浓度,优于其他组别(均P<0.05);②Western blot检测显示利拉鲁肽能够增强细胞p-Akt、Akt表达水平(均P<0.05),LY294002的加入能够降低p-Akt、Akt的表达(均P<0.05);③Brdu和划痕实验显示利拉鲁肽具有促进肺微血管内皮细胞增殖和迁移能力的作用(均P<0.05),而LY294002的加入能够降低上述细胞的增殖迁移能力(均P<0.05)。结论:利拉鲁肽对LPS诱导大鼠肺微血管内皮细胞的生物进程会产生显著影响,其机制可能与利拉鲁肽能够作用于PI3K/Akt信号通路有关。Objective:To explore the effect of liraglutide on the PI3K/Akt signaling pathway of rat pulmonary microvascular endothelial cells induced by lipopolysaccharide(LPS).Methods:The rat lung microvascular endothelial cells successfully cultured by LPS were used as experimental objects.Different concentrations of liraglutide(0,1,10,100,1000 nmol/L)were used to interfere cells.The cell growth curve was drawn by MTT method,and the effect of different concentrations of liraglutide on cell growth was analyzed.Western blot was used to detect the effects of normal cell,liraglutide(100 nmol/L)and PI3K/Akt pathway blocker(LY294002)on the expression of p-Akt and Akt proteins.Brdu fluorescent labeling method and scratch test were used to detect the effect of liraglutide on cell proliferation and migration.Results:By drawing the cell growth curve,it was found that 100 nmol/L liraglutide was the optimal cell growth concentration,which was better than other groups(all P<0.05).Western blot detection showed that liraglutide could enhance the expression level of Akt in cells,and the addition of LY294002 could reduce the expression of Akt(all P<0.05).Brdu and scratch experiments showed that liraglutide could promote the proliferation and migration of pulmonary microvascular endothelial cells and the addition of LY294002 could reduce the cell proliferation and migration ability(all P<0.05).Conclusion:Liraglutide has a significant effect on the biological process of rat lung microvascular endothelial cells induced by LPS.The mechanism may be related to the effect of liraglutide on the PI3K/Akt signaling pathway.
