摘 要:目的探讨一步法实时逆转录聚合酶链反应检测技术在手足口病病原学检测中的应用。方法对378例手足口病患者采集咽拭子(305份)、脑脊液(36份)、疱疹液(37份)标本,分别采用常规逆转录聚合酶链反应、一步法实时逆转录聚合酶链反应检测,比较两种方法检测EV71、CoxA16阳性率,比较一步法实时逆转录聚合酶链反应检测EV71、CoxA16及EV71+CoxA16阳性率。结果一步法实时逆转录聚合酶链反应检测EV71、CoxA16阳性率率显著高于常规逆转录聚合酶链反应(P<0.05)。EV71、CoxA16及EV71+CoxA16阳性率比较差异有统计学意义(P<0.01),结论一步法实时逆转录聚合酶链反应检测能提高手足口病患者病原学检测准确性,优于常规逆转录聚合酶链反应检测。Objective To explore the application of one-step real-time reverse transcription polymerase chain reaction(RT-PCR)detection technology in the etiology detection of hand-foot-and-mouth disease(HFMD).Methods The specimens of throat swabs(305 pieces),cerebrospinal fluid(36 pieces)and herpes fluid(37 pieces)from 378 HFMD patients were detected by routine RT-PCR and one-step real-time RT-PCR rapid detection.The positive rates of enterovirus 71(EV71)and coxsackievirus A16(CoxA16)by the two methods were compared.The positive rates of EV71,CoxA16 and EV71+CoxA16 by one-step real-time RT-PCR were compared.Results The positive rates of EV71 and CoxA16 by one-step real-time RT-PCR detection were significantly higher than those by routine RT-PCR(P<0.05).The differences in positive rates of EV71 and CoxA16 and EV71+CoxA16 were statistically significant(P<0.01).Conclusions One-step real-time RT-PCR detection can increase the accuracy of etiology detection in HFMD patients,which is better than routine RT-PCR.
