摘  要:目的观察荜茇细粉混悬对Beagle犬心血管系统和大、小鼠中枢神经系统的影响,评价荜茇的安全性。方法①Beagle犬单次灌胃给予荜茇细粉混悬液0.1、0.3和0.9 g·kg^(-1),利用植入式生理信号遥测系统(DSI)检测清醒Beagle犬给药前后的心电、血压指标,评价荜茇对Beagle犬心血管系统和体温的影响。②ICR小鼠单次灌胃给予荜茇细粉混悬液0.52、1.56和4.68 g·kg^(-1),观察小鼠自主活动情况和小鼠戊巴比妥钠阈下剂量催眠情况;SD大鼠单次灌胃给予荜茇细粉混悬液0.36、1.08和3.24 g·kg^(-1),观察大鼠协调运动情况,评价荜茇对大、小鼠中枢神经系统的影响。结果①与溶媒对照组相比,高剂量组Beagle犬给药2 h后心率降低,QT间期延长(P<0.05);荜茇细粉各剂量组对Beagle犬的血压和体温无明显影响。②与溶媒对照组比较,高剂量组小鼠给药30 min和1 h后活动次数均明显减少(P<0.01),其他各检测点活动次数未见差异;荜茇细粉各剂量组对小鼠睡眠潜伏期、入睡动物数、睡眠时间均无明显影响,证明荜茇细粉与戊巴比妥钠阈下剂量诱导小鼠睡眠无协同效应;荜茇细粉各剂量组对大鼠的协调运动潜伏期未见明显影响。结论本实验条件下,单次灌胃荜茇细粉对Beagle犬心血管系统和体温、对SD大鼠和ICR小鼠的中枢神经系统均未见明显影响。Objective To observe the effects of piper longum fine powder suspension on the cardiovascular system in Beagle dogs and the central nervous system in rats and mice,and evaluate the safety of piper longum.Methods①The Beagle dogs were intragastrically administered piper longum fine powder suspension once at 0.1,0.3 and 0.9 g·kg^(-1).The physiological signal telemetry system(DSI)was used to detect the ECG,blood pressure and body temperature of the sober Beagle dogs before and after the administration,and to evaluate the impact of piper longum on the cardiovascular system of Beagle dogs.②ICR mice were intragastrically administrered piper longum fine powder suspension once at 0.52,1.56 and 4.68 g·kg^(-1),and the spontaneous activity and the hypnosis effect of sub-threshold dose of pentobarbital sodium were observed in mice.SD rats were intragastrically administrered piper longum fine powder suspension once at 0.36,1.08 and 3.24 g·kg^(-1).The coordination function of the rats was observed and the effect of piper longum on the central nervous system in both rats and mice was evaluated.Results①Compared with the vehicle control group,the heart rate of Beagle dogs in the high-dose group was decreased and the QT interval was prolonged in 2 h after the administration(P<0.05).Various doses of piper longum fine powder had no significant effect on the blood pressure and body temperature in Beagle dogs.②Compared with the vehicle control group,activities of mice in the high dose group were also significantly reduced 30 min and 1 h after the administration(P<0.01).There was no significant effect in the activities at other test sites.Various doses of piper longum fine powder had no significant effect on the sleep latency,number of sleeping animals and the sleep time of mice,indicating no synergistic effect between piper longum fine powder and the sub-threshold dose hypnotic effect of sodium pentobarbital in mice.Each dose has no obvious effect on the coordination function of rats.Conclusion Under the experimental conditi
