摘 要:目的在复制功能性腹泻脾虚证大鼠模型后增加间断性高乳糖饮食的干预因素,观察参苓白术散对模型大鼠的治疗作用,并探讨其影响大鼠结肠水通道蛋白(Aquaporins,AQP)3、AQP8表达的机制研究。方法将56只大鼠随机分成正常组16只和模型组40只,模型组大鼠采用“高乳糖饮食喂养+水环境平台站立法”造模14 d;正常组大鼠在正常饲养环境下给予基本配方饲料喂养14 d。之后随机抽取正常组和模型组大鼠每组各8只,进行模型判定。其余32只模型组大鼠随机分成4组,分别为模型1组、模型2组、治疗1组和治疗2组。治疗1组和治疗2组给予参苓白术散治疗14 d;正常对照组、模型1组和模型2组给予蒸馏水灌胃14 d。给药期间模型1组和治疗1组大鼠在造模结束后的第4~7天和第11~14天给予高乳糖饲料喂养,其余时间给予基本配方饲料喂养;正常对照组、模型2组和治疗2组大鼠给予基本配方饲料。观察各组大鼠的一般情况,比较排便次数、腹泻指数、粪便含水率及小肠推进率,通过比色法检测血清乳糖含量、D-木糖含量以及淀粉酶活性,免疫荧光法观察结肠AQP3、AQP8的表达情况。结果与正常对照组比较,模型1组大鼠仍伴有神疲乏力,耳、舌、爪色淡白,毛发稀疏不泽,形体消瘦,纳少便溏,排便次数增加等症状,血清淀粉酶的活性下降(P<0.01),D-木糖含量降低(P<0.001),血清乳酸含量增加(P<0.01),小肠推进率增强(P<0.001),粪便含水率增加(P<0.001),结肠AQP3、AQP8表达水平显著降低(P<0.01),差异均有统计学意义。与模型1组比较,治疗1组大鼠精神状态等表征逐渐恢复,唇、舌、耳、鼻逐渐红润,毛发逐渐浓密柔亮,爪色恢复润泽,便溏乏力等症状消失,血清淀粉酶的活性增加(P<0.01),D-木糖含量增加(P<0.05),血清乳酸含量降低(P<0.05),小肠推进率降低(P<0.001),粪便含水率降低(P<0.001),结肠AQP3、AQP8表达水平升高(P<0.05),差异均Objective Intervening factors of intermittent high lactose diet were added after the rat model of functional diarrhea with spleen deficiency syndrome was cloned to observe the therapeutic effect of Shenling Baizhu Powder on the model rats,and to explore the mechanism of its influence on the expression of AQP3 and AQP8 in the rat colon.Methods A total of 56 rats were randomly divided into normal group(n=16)and model group(n=40).Model group rats were fed with high lactose diet and water environment platform legislation for 14 days.Rats in normal group were fed with basic formula feed for 14 days in normal feeding environment.Then,8 rats in each group were randomly selected from the normal group and the model group for model determination.The other 32 rats in model group were randomly divided into 4 groups,which were model group 1,model group 2,treatment group 1 and treatment group 2.Treatment group 1 and treatment group 2were treated with Shenling Baizhu powder for 14 days;Distilled water was given to normal control group,model group 1,model group 2 for 14 days.During administration,rats in the intermittent intervention group and the intermittent intervention group were given high lactose diet intervention on the 4 th to 7 th and 11 th to 14 th days after modeling,respectively,and the rest of the time were given basic formula feed feeding.Rats in normal control group,model group 2 and treatment group 2 were given basic formula feed.The general conditions of rats in each group were observed,and the defecation times,diarrhea index,fecal moisture content and small intestinal proptosis rate were compared.The serum lactose content,D-xylose content and amylase activity were detected by colorimetry,and the expression of AQP3 and AQP8 in colon was observed by immunofluorescence method.Results Compared with the normal control group,the rats in the model group 1 were still associated with fatigue,pale color of ear,tongue and paw,sparse hair,thin body,less diarrhea,increased defecation frequency and other symptoms,and decreas
