摘 要:目的探究谷红注射液有效成分羟基红花黄色素A(HSYA)介导内皮型一氧化氮合酶(eNOS)与凝素样氧化低密度脂蛋白受体(LOX-1)对人冠状动脉内皮细胞(HCAECs)损伤的影响。方法取HCAECs细胞采用加入20μg/ml氧化型低密度脂蛋白(ox-LDL)处理诱导细胞损伤模型作为模型组,将不同浓度(1、10、100μmol/L)HSYA处理过的细胞作为低、中、高HSYA组,将未处理的HCAECs细胞作为对照组,检测培养24 h各组细胞存活率及培养液中一氧化氮(NO)、乳酸脱氢酶(LDH)量,采用RT-PCR和Western blot检测各组LOX-1、eNOS的mRNA和蛋白表达量。结果CCK8检测结果显示,培养24 h时各HAYA处理组细胞存活率均高于模型组、低于对照组,且HAYA浓度越高细胞存活率越高,差异具有统计学意义(P<0.05);培养24 h时各HAYA处理组细胞培养液中NO含量均高于模型组、低于对照组,LDH含量低于模型组、高于对照组,且HAYA浓度越高NO含量越多,LDH含量越少,差异具有统计学意义(P<0.05);RT-PCR及Western blot检测结果显示,培养24 h时各HAYA处理组eNOS mRNA及蛋白相对表达量均高于模型组、低于对照组,LOX-1mRNA及蛋白表达量低于模型组、高于对照组,且HAYA浓度越高eNOS mRNA及蛋白表达量越高,LOX-1 mRNA及蛋白表达量越低,差异有统计学意义(P<0.05)。结论谷红注射液有效成分HSYA可呈剂量依赖性促进eNOS表达和抑制LOX-1蛋白表达,促进冠状动脉内皮细胞损伤修复。Objective To study the influence of endothelial nitric oxide synthase(eNOS)and lectin-like oxidized low-density lipoprotein receptor(LOX-1)mediated by hydroxysafflor yellow A(HSYA),an effective component in Guhong Injection,on the damage of human coronary artery endothelial cells(HCAECs).Methods The model of cellular damage was induced by adding oxidized low density lipoprotein(ox-LDL,20μg/ml)to HCAECs(model group).HCAECs were treated with HSYA in different doses(1μmol/L,10μmol/L,100μmol/L)and taken as low-dose HSYA group,mid-dose HSYA group and high-dose HSYA group.HCAECs without HSYA treatment were included into control group.The survival rate of HCAECs was detected after cultured for 24 h,and levels of nitric oxide(NO)and lactic dehydrogenase(LDH)in culture medium were detected.The mRNA and protein expressions of LOX-1 and eNOS were detected by using RT-PCR and Western blotting assay.Results The results of CCK8 detection showed that survival rate of HCAECs was higher in all HAYA groups than that in model group and lower than that in control group after culture for 24 h,and the higher the HAYA level,the higher the survival rate(P<0.05).The level of NO was higher in all HAYA groups than that in model group and lower than that in control group after culture for 24 h,LDH level was lower than that in model group and higher than that in control group,and the higher the HAYA level,the higher the NO level and the lower the LDH level(P<0.05).The results of RT-PCR and Western blotting assay showed that mRNA and protein expressions of eNOS were higher in all HAYA group than those in model group and lower than those in control group after cultured for 24 h,mRNA and protein expressions of LOX-1 were lower in all HAYA group than those in model group and higher than those in control group,and the higher the HAYA level,the higher the mRNA and protein expressions of eNOS and the lower the mRNA and protein expressions of LOX-1(P<0.05).Conclusion HSYA,an effective component in Guhong Injection,can improve eNOS expression and
