摘 要:目的探讨变应性鼻炎(AR)患者血清let-7e、miR-155-5p表达与Treg/Th17细胞平衡的关系。方法选择98例AR患者(AR组)和60例健康志愿者(对照组),采用实时荧光定量PCR法检测血清let-7e、miR-155-5p表达,流式细胞术检测Th17、Treg细胞,酶联免疫吸附试验检测血清IL-6、IL-17、IL-22、IL-23、IL-10、TGF-β1水平。分析let-7e、miR-155-5p表达与AR临床参数、Th17/Treg及其相关细胞因子间的关系。结果AR组let-7e表达、Treg细胞占比、Treg/Th17值、IL-10、TGF-β1低于对照组(P均<0.05),miR-155-5p表达、Th17细胞占比、IL-6、IL-17、IL-22、IL-23高于对照组(P均<0.05)。中重度、IgE阳性、伴哮喘患者血清let-7e表达低于轻度、IgE阳性、无哮喘患者(P均<0.05),miR-155-5p表达高于轻度、IgE阳性、无哮喘患者(P均<0.05)。相关性分析结果显示,血清let-7e与Treg、Treg/Th17、IL-10、TGF-β1呈正相关(r分别为0.406、0.467、0.435、0.467,P均<0.05),与Th17、IL-6、IL-17、IL-22、IL-23呈负相关(r分别为-0.512、-0.419、-0.464、-0.406、-0.312,P均<0.05),miR-155-5p与Treg、Treg/Th17、IL-10、TGF-β1呈负相关(r分别为-0.495、-0.468、-0.457、-0.368,P均<0.05),与Th17、IL-6、IL-17、IL-22、IL-23呈正相关(r分别为0.598、0.437、0.538、0.495、0.398,P均<0.05),let-7e与miR-155-5p呈负相关(r=-0.594,P<0.05)。结论AR患者血清let-7e表达下调,miR-155-5p表达上调,let-7e、miR-155-5p相互调节参与了ARTreg/Th17失衡过程。Objective To investigate the relationship between serum let-7E and miR-155-5P expression and Treg/Th17 cell balance in patients with allergic rhinitis(AR).Methods Ninety-eight AR patients(AR group)and 60 healthy volunteers(control group)were selected.The expression levels of serum Let-7E and miR-155-5P were detected by real-time fluorescence PCR(RT-PCR),and Th17 and Treg cells were detected by flow cytometry.Serum levels of inter‐leukin-6(IL-6),interleukin-17(IL-17),interleukin-22(IL-22),interleukin-23(IL-23),interleukin-10(IL-10)and transforming growth factor-β1(TGF-β1)were detected by enzyme chain immunosorbent assay.The relationship between let-7E and miR-155-5p expression and AR clinical parameters,Th17/Treg and related cytokines was analyzed.Results Let-7E expression,Treg cell proportion,Treg/Th17 ratio,IL-10,and TGF-β1 expression in the AR group were lower than those in the control group(all P<0.05),while the miR-155-5p expression,Th17 cell proportion,IL-6,IL-17,IL-22,and IL-23 were higher than those in the control group(all P<0.05).Serum let-7E expression in patients in the moderate to severe conditions,with IgE positive and asthma was lower than in patients in the mild state,with IgE positive and no asth‐ma(P<0.05),and miR-155-5p expression was higher than that in mild patients with IgE positive and no asthma(P<0.05).Correlation analysis results showed that serum let-7E was positively correlated with Treg,Treg/Th17,IL-10,and TGF-β1(r=0.406,0.467,0.435,and 0.467;all P<0.05),and was negatively correlated with Th17,IL-6,IL-17,IL-22,and IL-23(r=-0.512,-0.419,-0.464,-0.406,and-0.312;all P<0.05);miR-155-5p was negatively correlat‐ed with Treg,Treg/Th17,IL-10,and TGF-β1(r=-0.495,-0.468,-0.457,-0.368;all P<0.05),and was positively correlated with Th17,IL-6,IL-17,IL-22,and IL-23(r=0.598,0.437,0.538,0.495,and 0.398;all P<0.05).Let-7e was negatively correlated with miR-155-5p(r=-0.594,P<0.05).Conclusion Serum let-7E expression is down-reg‐ulated and the expression of miR-155-5p is up-regulated in AR pat
