摘 要:目的探究荧光PCR技术在手足口病病毒核酸检测中的应用价值。方法选取2017年8月至2019年12月本院收治的202例疑似手足口病患儿,所有患儿样本均采用荧光PCR技术、逆转录聚合酶链反应检测,比较两种检测方法下EV、EV-71、CA-16病毒阳性检出率及对手足口病的诊断情况。结果荧光PCR技术对EV、EV-71、CA-16病毒阳性检出率分别为55.45%、18.32%、33.66%,均高于逆转录聚合酶链反应的34.65%、5.94%、23.27%,EV+EV-71、EV+CA-16双阳性检出率为17.33%、32.67%,高于逆转录聚合酶链反应的9.90%、21.29%,差异有统计学意义(P<0.05);荧光PCR技术在手足口病中检出率为94.26%,高于逆转录聚合酶链反应的59.84%,差异有统计学意义(P<0.05)。结论采用荧光PCR技术诊断手足口病EV、EV-71、CA-16病毒阳性检出率较高,可为疾病诊断提供参考依据,以降低漏诊及误诊率。Objective To explore the application value of fluorescence PCR technology in nucleic acid testing of hand-foot-mouth disease virus.Methods 202 suspected cases of Hand-foot-mouth disease children admitted to our hospital from August 2017 to December 2019 were selected,all the children were detected by fluorescence PCR technology and reverse transcription polymerase chain reaction,and the positive detection rates of EV,EV-71,CA-16 virus and the diagnosis of hand-foot-mouth disease between the two detection methods were compared.Results The positive detection rates of EV,EV-71 and CA-16 virus by fluorescence PCR were 55.45%,18.32%and 33.66%,which were higher than 34.65%,5.94%,23.27%of reverse transcription polymerase chain reaction.The double positive detection rates of EV+EV-71 and EV+CA-16 were 17.33%and 32.67%,which were higher than the 9.90%and 21.29%of reverse transcription polymerase chain reaction,the difference was statistically significant(P<0.05);the detection rate of fluorescence PCR technology in Hand-foot-mouth disease was 94.26%,which was higher than 59.84%of reverse transcription polymerase chain reaction,the difference was statistically significant(P<0.05).Conclusion The use of fluorescent PCR technology in the clinical diagnosis of HEMD has high positive detection rate of EV,EV-71,and CA-16 viruses,which can provide a reference basis for disease diagnosis and reduce the rate of missed diagnosis and misdiagnosis.
