摘 要:为明确小麦质外体中葡萄糖浓度的变化与小麦条锈菌致病性的关系,以小麦条锈菌CYR32-杂46亲和组合与CYR32-贵农22非亲和组合为研究对象,分别提取2种小麦受侵染后的质外体溶液,采用酶法定量分析不同侵染组合的小麦质外体中葡萄糖浓度。结果表明,感病品种杂46在接种后0、1、2、3、4、8、14 d质外体中葡萄糖浓度呈现明显的波动,接种后0~3 d浓度急剧升高,3 d浓度达到峰值,为0 d浓度的21.23倍,接种后4 d浓度下降为峰值的0.32倍;抗病品种贵农22在接种后2 d质外体中葡萄糖浓度达到峰值,但仅为感病品种杂46浓度峰值的0.41倍;受到小麦条锈菌侵染后抗病品种比感病品种的质外体中葡萄糖浓度低并且差异显著。研究结果可为探究抗性小麦是否通过降低质外体中葡萄糖浓度限制Pst生长奠定基础。To investigate the relationship between concentrations of apoplastic glucose from wheat leaves infected by Puccinia striiformis f.sp.tritici(Pst)and the pathogenicity of Pst,the apoplastic fluid of wheat leaves in compatible interaction of CYR32-Hybrid 46 and incompatible interaction of CYR32-Guinong 22 were extracted,and apoplastic glucose concentrations were quantified by enzymatic method.The results showed that the apoplastic glucose concentrations of Hybrid 46 leaves fluctuated at 0,1,2,3,4,8,14 d post inoculation(dpi)after inoculation in compatible interaction between CYR32 and Hybrid 46.The glucose concentrations increased sharply from 0 to 3 dpi.The concentration increased 21.23-fold at 3 dpi compared with the glucose concentration at 0 dpi.The glucose concentration at 4 dpi declined to about 0.32-fold of the peak glucose concentration.The apoplastic glucose concentrations of Guinong 22 leaves in incompatible interaction rose to the highest value at 2 dpi,which was just 0.41-fold of the peak glucose concentration in compatible interaction.There was a significant decrease in apoplastic glucose concentrations from Guinong 22 compared with Hybrid 46 infected by Pst,which may provide a basis for exploring changes in apoplastic glucose concentrations from wheat linked to plant defense.
