摘 要:目的:探究姜黄素对科萨奇病毒B组3型(CVB3)诱导的病毒性心肌炎小鼠心肌损伤的保护作用及潜在的调节机制。方法:采用CVB3病毒诱导感染小鼠构建病毒性心肌炎模型。造模成功的小鼠随机均匀分为5个实验组,分别为模型组、环孢素组、姜黄素低剂量组、姜黄素中剂量组、姜黄素高剂量组,另以10只正常小鼠作为空白对照组,期间观察记录小鼠的一般状态及死亡情况;采用H&E染色观察小鼠心肌组织的病理学变化;采用ELISA法检测小鼠血清炎症相关因子TNF-α、IL-1β和IL-6及心肌损伤相关分子CK-MB和LDH含量变化;采用蛋白质免疫印迹法检测心肌组织炎症及凋亡相关分子Cleaved caspase-3、p-NF-κB和iNOS的表达情况。结果:CVB3病毒可成功诱导小鼠病毒性心肌炎组织学特征出现,姜黄素可呈剂量依赖性明显改善病毒感染模型小鼠心肌组织的破坏,横纹肌断裂溶解,纹理错乱等情况;与空白对照组比较,模型组小鼠血清炎症相关因子TNF-α、IL-1β和IL-6水平及心肌损伤相关分子CK-MB和LDH显著升高(均P<0.001)。与模型组比较,姜黄素低、中、高剂量组小鼠血清TNF-α、IL-1β和IL-6及CK-MB和LDH水平均明显降低(均P<0.001);与空白对照组比较,模型组小鼠心肌组织凋亡相关分子Cleaved caspase-3及炎症相关分子p-NF-κB和iNOS蛋白表达均显著上调(均P<0.001);与模型组小鼠比较,姜黄素低、中、高剂量组p-NF-κB、iNOS蛋白和姜黄素中、高剂量组Cleaved caspase-3蛋白表达均显著降低(均P<0.05)。结论:姜黄素可能通过抑制caspase-3、NF-κB和iNOS分子的蛋白表达改善CVB3病毒诱导的小鼠血清高TNF-α、IL-1β、IL-6、CK-MB和LDH水平。Objective:To explore the protective effect and potential regulatory mechanism of curcumin on myocardial injury induced by coxsackievirus type B3(CVB3)in mice with viral myocarditis.Methods:Viral myocarditismouse model was established by infected with CVB3 virus.The viral myocarditis mice were randomly divided into 5 groups:model group,cyclosporine group,low-dose curcumin group,middle-dose curcumin group and high-dose curcumin group.Ten normal mice were used as blank control group.The general state and death of mice were observed and recorded.The pathological changes of myocardium were observed by HE staining.The serum inflammatory factors TNF-α,IL-1βand IL-6 and myocardial injury related molecules CK-MB and LDH were detected by ELISA.The expressions of Cleaved caspase-3,p-NF-κB and iNOS were detected by Western blotting.Results:CVB3 virus induced the histological characteristics of viral myocarditis in mice,and curcumin significantly improved the myocardial tissue damage such as rupture and dissolution of striated muscle and texture disorder in virus-infected mice in a dose-dependent manner.Compared with the blank control group,the levels of TNF-α,IL-1β,IL-6,CK-MB and LDH in serum and the protein expressions of Cleaved caspase-3,p-NF-κB and iNOS in the model group were significantly elevated(P<0.001).Compared with the model group,the levels of serum TNF-α,IL-1β,IL-6,CK-MB and LDH in low-,medium-and high-dose curcumin groups were significantly decreased(P<0.001).The expressions of p-NF-κB and iNOS protein in low-,middleand high-dose curcumin groups and Cleaved caspase-3 protein in middle-and high-dose curcumin groups were significantly decreased(P<0.05).Conclusion:Curcumin may reverse CVB3 virus-induced high serum levels of TNF-α,IL-1β,IL-6,CK-MB and LDH in mice by down-regulating the expression of caspase-3,NF-κB and iNOS.
