摘 要:目的:研究黄芪多糖(APS)对心肌肥厚大鼠心肌组织中自噬表达的影响。方法:健康SD大鼠随机分为模型(AAC)组、假手术(Sham)组和APS组,AAC组及APS组均通过腹主动脉缩窄(AAC)术构建大鼠心肌肥厚模型,术后1周开始分别予灭菌蒸馏水、APS每天灌胃给药,连用8周。8周后测定各组大鼠心肌重量指数(HMI)、左室重量指数(LVMI);光镜下观察心肌组织学改变,Western blotting法检测心肌组织LC3Ⅱ/GAPDH、beclin-1的蛋白表达量,同时运用电子显微镜检测心肌细胞中自噬小体数量。结果:与Sham组相比,AAC组HMI、LVMI明显增加(P<0.01),光镜下观察到心肌纤维增粗、结构紊乱,心肌细胞直径和表面积均增加(P<0.01),LC3Ⅱ/GAPDH、beclin-1表达水平明显升高(P<0.05),电子显微镜下可见自噬小体明显增加(P<0.01);与AAC组相比,APS组HMI、LVMI显著降低(P<0.01);苏木精-伊红(HE)染色可见心肌细胞肥大程度较AAC组减轻,心肌纤维排列整齐;心肌细胞直径和表面积明显减小;APS组LC3Ⅱ/GAPDH、beclin-1表达水平均有不同程度下降(P<0.05),电镜下可见自噬小体明显减少。结论:APS在大鼠压力超负荷心肌肥厚中起保护作用,其机制可能为通过调控自噬水平实现的。Objective:to study the protective effect of Astragalus Polysaccharide(APS)on myocardial hypertrophy in rats and its underlying mechanism.Methods:the rat model of myocardial hypertrophy was established by abdominal aortic coarctation(AAC).SD rats were randomly divided into model group(AAC),sham operation group(sham)and APS group.After 8 weeks,the myocardial weight index(HMI),left ventricular weight index(LVMI),myocardial histology and the protein expression of LC3Ⅱ/GAPDH and beclin-1 in myocardial tissue were measured,and the number of autophagosomes in myocardial cells was detected by electron microscope.Results:compared with sham group,the levels of HMI and LVMI in AAC group were increased(P<0.01),the diameter and surface area of myocardial cells were increased(P<0.01),the expression level of LC3Ⅱ/GAPDH was significantly increased,and the number of autophagosomes was significantly increased(P<0.05);compared with AAC group,the levels of HMI and LVMI in APS group were significantly decreased Compared with AAC group,he staining showed that the degree of hypertrophy of cardiomyocytes was reduced,and the arrangement of myocardial fibers was orderly;the diameter and surface area of cardiomyocytes were significantly reduced.The expression levels of LC3Ⅱ/GAPDH and beclin-1 were decreased in APS group,and auto-phagosomes were significantly decreased under electron microscope.Conclusion:Astragalus polysaccharide plays a protective role inmyocardial hypertrophy of ratscaused by stress overload,and its regulation of autophagy may be involved in the inhibition of Astragalus Polysaccharide on myocardial hypertrophy.
