摘 要:目的探讨维甲酸受体在TmLRP术后修复中的作用。方法2018年2月至2018年10月,比格犬(购自南京医科大学动物中心)按照体重顺序随机分为4组,每组3只,1组为对照组。3个实验组分别行铥激光前列腺切除术。分别于手术后1、2、3周处死犬,并完整切取整个前列腺组织。免疫组织化学及免疫荧光检查3个维甲酸受体亚型(RAR)表达分布。进一步定量PCR、蛋白质印迹法(Western blot)定量检测3个受体亚型。SAS软件非参数检验统计数据。结果维甲酸受体三个亚型(RARα、RARβ、RARγ)存在于正常的比格犬前列腺组织中,铥激光前列腺术后创面组织中RARα和RARγ术后表达逐渐增加,qPCR显示RARβ术后1周增加明显(6.63±0.50,t=29.820,P<0.01),而后逐渐下降第2周(4.29±0.22,t=15.560,P<0.01),第3周(2.31±0.41,t=12.270,P<0.01)。免疫荧光显示RARβ表达于创面间质组织的巨噬细胞中。结论巨噬细胞可能通过RARβ作用于间质细胞参与铥激光前列腺术后创面修复过程。Objective The process of wound healing is important for the reduction of thulium laser prostatectomy(TmlRP)postoperative complications.The possible role of retinoic acid receptor(RAR)in the process of wound healing was studied.Methods The study period was from February 2018 to October 2018.Beagles were divided into 4 groups with 3 beagles in each group and 1 group served as the control group.After TmlRP,the prostate tissues were removed at 1st,2nd and 3rd week after surgery.Immunohistochemistry and immunofluorescence were used to detect the expression and distribution of three RAR subtypes.They were further quantitatively detected by real-time PCR and Western blotting.Results The three subtypes were present in normal beagle prostate tissues.The expression levels of RARαand RARγin the wound tissues were gradually increased.The qPCR showed that RARβincreased significantly(6.63±0.50,t=29.820,n=9,P<0.01)1 week after operation,and then decreased gradually(4.29±0.22,t=15.560,n=9,P<0.01)2 weeks after operation and(2.31±0.41,t=12.270,n=9,P<0.01)3 weeks after operation.RARβwas expressed in macrophages in the wound mesenchymal tissue,and there was a trade-off between macrophages and mesenchymal cells.Conclusion Macrophages may participate in the wound repair process after TmlRP by acting on the mesenchymal cells through RARβ.
