摘 要:目的研究复方丹参制剂中丹酚酸B、丹参素、丹参酮Ⅰ、丹参酮ⅡA、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1在大鼠血浆中的药动学。方法LC⁃MS/MS分析采用XBridge BEH C_(18)色谱柱(1.7μm,2.1 mm×100 mm);流动相水(含0.1%甲酸)⁃甲醇,梯度洗脱;体积流量0.3 mL/min;柱温40℃;电喷雾离子源(ESI);多反应监测(MRM)模式。通过DAS3.0软件计算药动学参数。结果7种成分在各自范围内线性关系良好(r>0.9900),基质效应82.9%~124.0%,提取回收率72.1%~93.8%。结论复方丹参制剂中丹参素、人参皂苷Rb1是主要入血成分。人参皂苷Rb1、丹参酮Ⅰ体内消除较慢,而三七皂苷R1较快。AIM To study the pharmacokinetics of salvianolic acid B,danshensu,tanshinoneⅠ,tanshinoneⅡA,notoginsenoside R1,ginsenoside Rg1 and ginsenoside Rb1 in Compound Danshen Preparations in rat plasma.METHODS The LC⁃MS/MS analysis was performed on a 40℃thermostatic XBridge BEH C18column(1.7μm,2.1 mm×100 mm),with the mobile phase comprising of water(containing 0.1%formic acid)⁃methanol flowing at 0.3 mL/min in a gradient elution manner.Electrospray ionization(ESI)was adopted in multiple reaction monitoring(MRM)mode.The pharmacokinetic parameters were calculated by DAS3.0 software.RESULTS Seven constituents showed good linear relationships within their own ranges(r>0.9900),whose matrix effects were 82.9%-124.0%with the extraction recoveries of 72.1%-93.8%.CONCLUSION Danshensu and ginsenoside Rb1 are main constituents migrating to blood in Compound Danshen Preparations.Ginsenoside Rb1 and tanshinoneⅠdemonstrate slowly in vivo elimination,while that of notoginsenoside R1 is rapid.
