摘 要:目的:通过星点设计-效应面法优选出黄精固体饮品中黄精多糖成分的最佳提取工艺。方法:采用水煎煮法提取黄精多糖成分,用蒽酮-硫酸法测定黄精多糖含量。以料液比和提取时间为考察因素,黄精多糖含量和干膏率作为评价指标,进行二项式拟合建立模型,从二项式描绘三维效应面分析得到总评归一值(OD)较佳的实验条件,按照优化出的最佳提取条件进行验证研究。结果:通过采用星点-设计效应面法得到最佳模型,最佳提取工艺为:料液比为1∶11,提取时间100min,提取2次。三次平行验证试验得到黄精多糖平均含量606.23mg/g、平均干膏率0.498%,总评OD值与理论预测值的偏差为2.92%。结论:星点设计-效应面法结合蒽酮-硫酸法优选黄精固体饮品中黄精多糖成分的最佳水提取工艺可行,为后续的黄精固体饮品大生产提供参考。Objective:To optimize the optimal extraction process for Polygonatum sibiricum polysaccharide in rhizoma Polygonatum sibiricum solid beverage.Methods:The components of Polygonatum sibiricum polysaccharide were extracted by water decoction,and the content of Polysaccharide from rhizoma Polygonatum was determined by anthrone-sulfuric acid method.Taking solid-liquid ratio and extraction time as the investigation factors,the content of polysaccharide and the dry paste rate of rhizoma Polygonatum were used as evaluation indexes,and the binomial fitting was used to establish the model.From the analysis of the three-dimensional effect surface analysis,the total evaluation value(OD)was better.The experimental conditions were verified by the optimized extraction conditions.Results:The best model was obtained by using the star point-design effect surface method.The optimal extraction process was as follows:The solid liquid ratio was 1∶11,extraction time-length was100 min,extraction time was 2 times.Three parallel verification tests showed that the average content of Polygonatum sibiricum polysaccharides was 606.23mg/g,the average dry paste rate was 0.498%,and the deviation of the overall evaluation OD value from the theoretical prediction value was 2.92%.Conclusion:Central composite design response surface method combined with anthrone-sulfuric acid method is the best water extraction process for Polygonatum sibiricum polysaccharide in rhizoma Polygonatum solid beverages.
