摘  要:在PDA培养基中添加NH4Cl和在灭菌发酵料中添加NH4OH(氨水)分析糙皮侧耳(Pleurotus ostreatus)不同菌株黑抗650、野生株NY-2和黑平17-1对氨的耐受性,并通过在NH4Cl质量浓度为1.00%和1.25%的PDA培养基中共培养(32℃)考察1-氨基环丙烷-1-羧酸脱氨酶产生菌对糙皮侧耳氨耐受性的影响。结果表明:在PDA培养基中培养,糙皮侧耳黑抗650、野生株NY-2和黑平17-1菌丝生长的适宜NH4Cl质量浓度分别为0.10%~0.20%、0.10%~0.20%和0.75%,生长上限质量浓度均为0.75%;在灭菌的糙皮侧耳发酵料中培养,3个糙皮侧耳菌株菌丝生长的适宜氨质量浓度为0.08%~0.25%,生长上限质量浓度均为0.25%。将1-氨基环丙烷-1-羧酸脱氨酶产生菌假单胞菌(Pseudomonas sp.)UW4与黑抗650共培养,可提高糙皮侧耳菌丝在NH4Cl质量浓度为1.00%和1.25%的PDA培养基中生长速度,分别提高13.82%和32.84%。糙皮侧耳对氨有较高的耐受性,1-氨基环丙烷-1-羧酸脱氨酶产生菌能够促进糙皮侧耳菌丝在发酵料高温和高氨条件下生长。Different strains of Pleurotus ostreatus,Heikang 650,wild-type NY-2,Heiping 17-1were analyzed for their tolerance to ammonia by adding NH4Cl and NH4OH to PDA medium and sterilized compost,respectively.In addition,1-aminocyclopropane-1-carboxylic acid deaminase(AcdS)producing bacterium Pseudomonas sp.UW4and its acdS gene deletion mutant were co-cultured with Heikang 650on PDA containing either 1.00%or 1.25%of NH4Cl at 32℃.The results showed that the optimal NH4Cl concentration in PDA for Heikang 650,wild strain NY-2and Heiping 17-1were 0.10%-0.20%,0.10%-0.20%and 0.75%,respectively.The upper-limit NH4Cl concentration for all three strains was 0.75%.The optimal range of NH4OH concentration in the sterilized compost for all three strains was 0.08%-0.25%,and the upper-limit concentration was 0.25%.Co-culture of Heikang 650together with UW4increased the mycelial growth rate by 13.82%and 32.84%at 1.00%and 1.25%NH4Cl in PDA,respectively.The results suggested that P.ostreatus had high tolerance to ammonia,and Pseudomonas sp.UW4 promoted P.ostreatus mycelial growth under high temperature and high ammonia concentrations.
