摘  要:目的筛选一种操作简单、分辨率高的烟曲霉基因分型方法。方法选取分离自14例慢性肺曲霉病(CPA)患者的31株烟曲霉,同时用两种基于串联重复序列[改良烟曲霉短串联重复序列(改良STRAf)和表面蛋白编码基因外显子的串联重复序列(TRESP)]的方法进行基因分型,用NCSS 11软件绘制树状图,比较两种方法的分型效果。结果改良STRAf方法共鉴定出23种基因型,其中MC3是分辨率最高的基因,共鉴定出14个等位基因;TRESP方法共鉴定出18种基因型,MP2、CSP、CFEM最常见的基因型分别为m1.1、t03、c08A,其中MP2是分辨率最高的基因,共鉴定出8个等位基因;菌株11、13的改良STRAf分型结果一致,但CSP基因存在一个短串联重复序列(STR)差别;菌株23与M患者分离菌株的TRESP分型一致,但其改良STRAf方法多个基因的STR数存在差别。结论 TRESP的分辨率略低于改良STRAf方法,但其不依赖于仪器,适合在临床实验室推广使用。Objective To select a simple and highly discriminating method for genotyping Aspergillus fumigatus. Methods A total of 31 A. fumigatus isolates from 14 chronic pulmonary aspergillosis(CPA)patients were chosen and genotyped by both short tandem repeat(STR)-based methods:modified short tandem repeats of A. fumigatus(mSTRAf)and tandem repeats within exons of surface protein coding genes(TRESP). Dendrogram was created by NCSS 11 software and the discriminatory power of the two methods was compared. Results A total of 23 and 18 genotypes were identified by mSTRAf and TRESP,respectively. The highest discriminatory power was seen in MC3 and MP2 gene. A total of 14 and 8 alleles were identified in MC3 and MP2 gene,respectively. The most common allele in MP2,CSP and CFEM were m1.1,t03,and c08 A,respectively. The identical mSTRAf genotype,but one STR difference in CSP was seen in isolate 11 and 13. Isolate23 shared the same TRESP profile with the isolates from patient M,but differed in mSTRAf STR numbers. Conclusions The discriminatory power of TRESP method was slightly lower than the modified STRAf. However,TRESP doesn’t require for the instrument and is suitable for the clinical laboratory.
