摘 要:目的探讨妊娠妇女炎性阴道分泌物中菌群结构与白细胞浸润数量的相关性,为临床女性泌尿生殖系统疾病研究提供可靠的理论依据。方法运用高通量二代测序技术检测20例(其中1+WBC 3例,2+WBC 6例,3+WBC10例,4+WBC 1例)妊娠妇女不同程度炎性阴道分泌物中微生物;采用秩和检验和卡方检验进行个体间显著性差异分析,使用生物信息学软件进行数据处理。结果 20份样本16S r RNA和ITS分析共注释出9个门、8个纲、8个目、9个科、8个属和9个种水平物种。16S rRNA分析中Simpson指数显示不同样本间物种多样性差异不显著,无统计学意义(P>0.05)。妊娠妇女不同程度炎性阴道分泌物中菌群结构差异不显著。妊娠妇女在种水平仅有德氏乳杆菌、瑞氏乳杆菌、惰性乳杆菌、罗伊乳杆菌、Atopobium vaginae、Campylobacter ureolyticus、人阴道乳杆菌、白假丝酵母和近平滑假丝酵母共9个物种。结论妊娠妇女炎性阴道分泌物中菌群结构不随白细胞数量的增加而改变,个体间菌群分布比较相似且稳定。Objective To explore the correlation between the bacterial flora structure and the number of white blood cell(WBC)infiltration in inflammatory vaginal secretions of pregnant women,and to provide a reliable theoretical basis for the clinical study of female genitourinary system diseases. Methods High-throughput second-generation sequencing technology was used to detect microorganisms in different degrees of inflammatory vaginal secretions in 20 pregnant women,there were3 cases of 1+WBC,6 cases of 2+WBC,10 cases of 3+WBC,and 1 case of 4+WBC.Rank-sum test and chi-square test were used to analyze the significance difference among individuals.Bioinformatics software was used for data processing.Results A total of 9 phyla,8 classes,8 orders,9 families,8 genera and 9 species were identified by 16 S ribosomal variable region and fungal internal transcribed spacer region of 20 samples.Simpson index of 16 S rRNA analysis showed that there was no significant difference in species diversity among different samples(P>0.05). There was no significant difference in bacterial flora structure in different degrees of inflammatory vaginal secretions of pregnant women. There were only 9 species of Lactobacillus delbrueckii,Lactobacillus helveticus,Lactobacillus inert,Lactobacillus reuteri,Atopobium vaginae,Campylobacter ureolyticus,Lactobacillus coleohominis,Candida albicans and Candida parapsilosis in the vaginal secretions of pregnant women. Conclusion The bacterial flora structure in inflammatory vaginal secretions of pregnant women did not change with the increase of white blood cell number,and the distribution of bacterial flora between individuals was similar and stable.
