摘 要:目的探究电针心包经穴对大鼠脑缺血后神经修复的作用机制。方法将SD大鼠随机分为正常组、假手术组、模型组、心包经组、肺经组,每组6只。采用颈外动脉插入线栓法制造局灶性大脑中动脉缺血模型。大陵、内关、曲泽、天泉四穴为心包经组取穴,太渊、列缺、尺泽、天府四穴为肺经组取穴,每日电针30 min,连续3 d。通过苏木精-伊红(HE)染色观察大鼠缺血脑组织病理形态学改变,酶联免疫吸附法(ELISA)检测血清中神经生长抑制因子(Nogo-A),Ras同源基因家族成员A(RhoA),Rho激酶Ⅱ(ROCKⅡ)含量,实时荧光定量PCR法(RT-qPCR)检测脑梗死区Nogo-A、NgR1、RhoA、和ROCKⅡmRNA的表达。结果造模后模型组大鼠脑组织中出现大片梗死灶,神经细胞大片消失或细胞皱缩;电针心包经和肺经穴能有效改善脑组织结构和细胞形态,且心包经组优于肺经组。与正常组、假手术组相比,模型组大鼠血清中Nogo-A、RhoA、RockⅡ含量均上升(P<0.01),缺血侧脑组织中Nogo-A、NgR1、RhoA及RockⅡmRNA表达均显著升高(P<0.05);与模型组比较,心包经组血清中Nogo-A、RhoA含量和脑组织中Nogo-A、NgR1、RhoA mRNA的表达均下降(P<0.05),肺经组脑组织中NgR1、RhoA mRNA的表达降低(P<0.01);与肺经组相比,心包经组血清中Nogo-A含量下降(P<0.01),脑组织中NgR1、RhoA mRNA的表达下降(P<0.01)。结论电针心包经穴能改善脑组织结构和细胞形态并通过下调神经生长抑制信号通路Nogo-A/NgR1-RhoA/RockⅡ中关键分子基因的表达促进脑缺血后神经功能修复。Objective To explore the mechanism in nerve repair after cerebral ischemia with electroacupuncture at Pericardium Meridian points in rat models.Method The SD rats were randomized into a normal group,a sham operation group,a model group,a Pericardium Meridian group and a Lung Meridian group,with 6 rats in each group.The middle cerebral artery ischemia model was established using external carotid artery occlusion method.Daling(PC7),Neiguan(PC6),Quze(PC3)and Tianquan(PC2)were selected in the Pericardium Meridian group,and Taiyuan(LU9),Lieque(LU7),Chize(LU5)and Tianfu(LU3)were selected in the Lung Meridian group,for electroacupuncture intervention,30 min a day for 3 consecutive days.Histopathologic change in rat’s ischemic brain tissue was observed using HE staining;ELISA was employed to detect the contents of nerve growth inhibitor Nogo-A,Ras homolog gene family member A(RhoA),Rho-associated protein kinaseⅡ(ROCKⅡ)in serum;RT-qPCR was used to determine the mRNA expressions of Nogo-A,NgR1,RhoA and ROCKⅡin the infarction area.Result After modeling,massive infarction occurred in the rats’brain tissue,along with extensive loss or shrinkage of nerve cells;electroacupuncture at Pericardium Meridian and Lung Meridian acupoints both can effectively improve the structure of brain tissue and morphology of brain cells,and the Pericardium Meridian group was superior to the Lung Meridian group.Compared with the normal and sham operation groups,the contents of serum Nogo-A,RhoA and ROCKⅡincreased in the model group(P<0.01),and the mRNA expressions of Nogo-A,NgR1,RhoA and ROCKⅡincreased notably in the ischemic brain tissue(P<0.05);compared with the model group,the contents of serum Nogo-A and RhoA in serum and the mRNA expressions of Nogo-A,NgR1 and RhoA in brain tissue declined in the Pericardium Meridian group(P<0.05),so did the mRNA expressions of NgR1 and RhoA in brain tissue in the Lung Meridian group(P<0.01);compared with the Lung Meridian group,the content of Nogo-A in serum dropped(P<0.01)and the mRNA expressi
