摘  要:目的研究心肌梗死大鼠心肌组织中血管生成素2(Ang2)和蛋白激酶B(AKT)的基因和蛋白表达及动态变化,探索心肌梗死后血管新生的信号通路。方法健康清洁级雄性SD大鼠24只,随机分为假手术组(sham组)和心肌梗死组(MI组),通过结扎左冠状动脉前降支(LAD)建立心肌梗死模型,假手术组只穿线不结扎。分别于术后7 d、14 d两个时间点,每组随机取6只处死,留取心脏左心室前壁同一部位心肌组织,切片行2,3,5-氯化三苯基四氮唑染色(TTC)染色,观察模型建立成功与否。聚合酶链反应(PCR)法检测Ang2、AKT基因表达水平,蛋白质印迹法检测Ang2、总AKT和磷酸化AKT的蛋白表达。结果①TTC染色结果提示:sham组大鼠前降支供血区心肌组织呈红色;MI组大鼠结扎线以下心肌组织呈粉白色;②PCR结果示:MI组Ang2、AKT的mRNA表达较sham组明显增加(P<0.05);14 d与7 d组相比Ang2、AKT的mRNA表达明显增加(P<0.05);③蛋白质印迹结果提示:MI组Ang2、AKT和磷酸化的AKT蛋白表达较sham组明显增加(P<0.05);14 d与7 d组相比,MI组Ang2和磷酸化的AKT蛋白表达均增加(P<0.05),而总AKT蛋白表达差异无统计学意义,sham组的Ang2、总AKT和磷酸化的AKT蛋白表达差异均无统计学意义。相关性分析结果提示Ang2和磷酸化的AKT蛋白表达呈正相关。结论Ang2可能通过AKT途径发挥促心肌梗死后血管生成作用,且这一过程可能是通过诱导AKT磷酸化实现的。Objective To investigate the mRNA and protein expression and dynamic changes of Ang2 and AKT in myocardial tissue of rats with myocardial infarction,and to explore the signal pathway of angiogenesis after myocardial infarction.Methods Twenty-four healthy and clean male SD rats were randomly divided into sham operation group(sham group)and myocardial infarction group(MI group).Myocardial infarction was modeled by ligating the left anterior descending coronary artery(LAD).The sham group received threading without ligation.At 7d and 14 d postoperatively,six rats in each group were randomly selected and sacrificed.The myocardial tissues were sampled from a same part of anterior wall of the left ventricle,sectioned and stained with TTC,to validate the success of modelling.PCR was used to detect the mRNA expression of Ang2 and AKT,and Western botting was used to examine the protein expression of Ang2,total AKT and phosphorylated AKT.Results①After TTC staining,the myocardial tissue supplied by anterior descending branch in the sham group appeared red,while those below the ligature in the MI group appeared pink to pale in color.②PCR showed that the mRNA expression levels of Ang2 and AKT in the MI group was significantly higher than those in the sham group(P<0.05),and were also significantly higher at 14 d compared with those at 7 d(P<0.05).③Western blotting showed that the protein expression levels of Ang2,total AKT and phosphorylated AKT in the MI group was significantly higher than those in the sham group(P<0.05).Compared with 7 d,the protein expression levels of Ang2 and phosphorylated AKT in the MI group were significantly higher at 14 d(P<0.05),while the difference in total AKT protein expression was not statistically significant.The protein expression levels of Ang2,total AKT and phosphorylated AKT in the sham group did not differ significantly between 7 d and 14 d.Correlation analysis suggested a significant correlation between the Ang2 and phosphorylated AKT protein expression.Conclusion Ang2 may promote an
