摘 要:目的建立同时测定雷公藤中雷公藤内酯甲和雷公藤甲素含量的分析方法,比较不同炮制方法对雷公藤药材中雷公藤内酯甲和雷公藤甲素含量的影响及炮制前后的变化。方法采用外标法,以0.1%磷酸乙腈(A)-0.2%三乙胺水溶液(B)为流动相,梯度洗脱;检测波长为210 nm;流速为1.0 mL/min。结果雷公藤内酯甲和雷公藤甲素分别在7.13~285.20 μg/mL(r=0.9990)和6.13~245.20μg/mL(r=0.9999)范围内呈良好的线性关系,加样回收率为96.24%~104.53%,RSD为0.9%~2.7%。不同炮制方法的雷公藤药材中雷公藤内酯甲和雷公藤甲素的含量差异显著,其中酒炙方法对上述两种成分影响较大。结论本实验揭示炮制方法对雷公藤的雷公藤甲素和雷公藤内酯甲的含量有一定的影响,为后续雷公藤减毒增效研究提供基础资料。Objective To establish a high performance liquid chromatographic method for the determination of wilforlide A and triptolide in Tripterygium wilfordii,and investigate the influence of wilforlide A and triptolide of the different processing Methods of Tripterygium wilfordii.Methods Using external standard method,the mobile phase was acetonitrile(0.1%phosphoric acid,A)-triethylamine(0.2%,B),the detection wavelength was 210 nm,and the flow rate was 1.0 mL/min.Results Wilforlide A and TRiptolide had a good linearity in the range of 7.13~285.20μg/mL(r=0.9990)and 6.13~245.20μg/mL(r=0.9999)respectively.The average recoveries of standard addition for the compounds were between 96.24%and 104.53%The different processing methods of Tripterygium wilfordii.had certain influence to 2 kinds of components.The wine processing method has a great influence on the above two components.Conclusion This experiment revealed that the processing method had a certain influence on the main components of Tripterygium wilfordii.,to provide the basic data for the follow-up study on the toxicity and synergism of Tripterygium wilfordii.
