摘 要:采用RNAseq测序技术对比变异轴孔珊瑚(Acropora valida)在酸化高温胁迫及去胁迫条件下的mRNA转录组。构建了12个cDNA文库,共获得451789689个双端片段。通过拼接共得到1 056 727个转录本,其中86.5%匹配到珊瑚序列。转录本的差异表达分析显示,胁迫条件下共6 328个转录本发生了显著变化,主要涉及热休克蛋白、物质转运蛋白、凋亡调节和钙化四个方面。其中98.1%的差异表达转录本在胁迫去除后都能恢复到对照组水平;结果表明,尽管变异轴孔珊瑚在酸化高温胁迫时受到很大影响,但其转录本表达在胁迫去除后能恢复到原来的水平。首次探究酸化高温双胁迫去除后变异轴孔珊瑚转录本表达水平的恢复潜力,对研究珊瑚如何应对全球气候变化有一定指导意义。RNAseq sequencing technology was performed to compare the mRNA expression of Acropora valida under acidic-thermal stress and after the stress is lifted.Twelve cDNA libraries were constructed,and a total of 451789689 double-ended fragments were obtained.A total of 1056727 contigs were obtained after de novo assembling.Through differential expression analysis of the A.valida contigs,it was found that a total of 6328 contigs were significantly changed under stress conditions,which include contigs encoding heat shock proteins,substance transporters,apoptosis,and calcification related proteins;and 98.1%of the contigs were recovered after stress release.A.valida was obviously affected by acidification and high temperature stress,but its gene expression could be recovered to original level after stress removal.This study is the first to explore the potential for restoration of the transcript expression level of the corals under and after dual acidic-thermal stress,and has certain guiding significance for studying how corals respond to global climate change.
