摘 要:试验旨在研究日粮纤维水平对苏禽3号肉鸡盲肠纤维素酶活性、肠道组织形态及生长相关基因表达的影响。选择体重相近的苏禽3号肉鸡360只,随机分为3组,每组4个重复。3组分别为2%日粮纤维(对照组)、4%日粮纤维组(试验1组)、6%日粮纤维组(试验2组)。试验期为10 w。采集盲肠内容物测定纤维素酶活性,观察十二指肠、空肠、回肠和盲肠组织形态,检测小肠中相关生长发育基因表达量。结果表明:日粮纤维添加可显著提高盲肠内容物纤维素酶活性(P<0.05)。回肠和空肠黏膜层厚度随着日粮纤维水平提高显著提高(P<0.05)。随着日粮纤维水平的提高,各基因的表达量都呈现先上升后下降的趋势,其中日粮纤维水平在4%时,BMP4基因、SMAD4基因和GATA3基因在小肠各段中表达量最高。研究表明,日粮纤维水平提高可以提高盲肠内容物纤维素酶活性,显著影响苏禽3号肉鸡肠道组织形态,促进相关生长发育基因的表达。The purpose of the experiment was to study the effect of dietary fiber levels on the cecal cellulase activity,intestinal tissue morphology and growth-related gene expression of Suqin No.3 broilers.Totally 360 Suqin No.3 broilers with similar body weight were randomly divided into three groups with four replicates in each group.They were divided into 2% dietary fiber(control group),4%dietary fiber group(group one)and 6% dietary fiber group(group two).The experiment lasted for 10 w.Cecal contents were collected to determine the cellulase activity,the morphology of duodenum,jejunum,ileum and cecum were observed,and the expression levels of growth-related genes were detected in small intestine.The results showed that dietary fiber supplementation could significantly increase the cellulase activity of cecal contents(P<0.05).With the increase of dietary fiber level,the villus height and crypt depth of duodenum,jejunum and ileum were not significantly different(P<0.05).The thickness of ileum and jejunum mucosa was also increased significantly with the increase of dietary fiber level.With the increase of dietary fiber level,the expression levels of BMP4 gene,SMAD4 gene and GATA3 gene were the highest in all segments of small intestine when dietary fiber level was 4%.The results showed that the increase of dietary fiber level could increase the cellulase activity of cecal contents,significantly affect the intestinal morphology of Suqin No.3 broilers,and promote the expression of related growth and development genes.
