摘 要:目的:研究低强度交变电磁场(electromagnetic fields,EMF)治疗对于丙泊酚诱发老年大鼠认知功能衰退的作用及机制。方法:选取36只20月龄雄性Sprague-Dawley老年大鼠,随机分为丙泊酚组、丙泊酚+EMF组以及对照组(无丙泊酚及EMF干预),每组12只。丙泊酚组和丙泊酚+EMF组腹腔注射丙泊酚,首次注射剂量为60 mg/kg,随后每小时注射1次丙泊酚(注射剂量为30 mg/kg),共注射6次(即可维持麻醉状态6 h)。丙泊酚+EMF组大鼠每天进行6 h的全身EMF暴露(5 mT、50 Hz),连续7 d。使用水迷宫测试学习记忆功能,使用酶联免疫吸附测定法检测血清淀粉样β蛋白(Aβ)、血清神经元特异性烯醇化酶(NSE)、海马神经生长因子(NGF)及脑源性神经营养因子(BDNF)表达水平,分别使用TUNEL染色和蛋白质免疫印迹分析海马组织细胞凋亡水平及抗凋亡因子Bcl-2、凋亡标记物caspase-3表达水平。采用SPSS 20.0软件进行统计分析。结果:与丙泊酚组相比,丙泊酚+EMF组大鼠的逃逸潜伏期和游泳距离显著减少(P<0.05),穿越平台次数显著增加(P<0.05),血清Aβ和NSE表达水平显著降低(P<0.05),海马组织NGF和BDNF表达水平显著升高(P<0.05),海马凋亡细胞数量以及caspase-3的表达水平显著降低(P<0.05),Bcl-2表达水平显著升高(P<0.05)。丙泊酚+EMF组大鼠与对照组相比,在逃逸潜伏期、游泳距离、穿越平台次数以及Aβ、NSE、NGF、BDNF、Bcl-2、caspase-3表达水平方面均无统计学差异(P>0.05)。结论:低强度交变EMF能够显著改善丙泊酚诱发老年大鼠认知功能衰退症状,这与其对细胞凋亡的抑制以及Aβ、NSE、NGF和BDNF的调控相关。Objective:To study the effect and mechanism of low-intensity alternating electromagnetic field(EMF)on propofol-induced decline of the cognitive function in aging rats.Methods Thirty six 20-month-old male Sprague Dawley rats were randomly divided into a propofol group,a propofol+EMF group and a blank control group(with no propofol or EMF stimulation)with 12 rats in each group.In the propofol and propofol+EMF groups,propofol was injected intraperitoneally at an initial dose of 60 mg/kg,followed by one injection of propofol(at a dose of 30 mg/kg)every hour for a total of six injections(i.e.,to maintain anesthesia for 6 h).The rats in the propofol+EMF group were exposed to whole-body EMF(5 mT,50 Hz)for6 h daily for 7 d.Learning memory function was tested using the water maze,and serum amyloidβprotein(Aβ),serum neuronspecific enolase(NSE),hippocampal nerve growth factor(NGF)and brain-derived neurotrophic factor(BDNF)expression levels were measured using enzyme-linked immunosorbent assay.The levels of apoptosis,anti-apoptotic factor Bcl-2 and apoptotic marker caspase-3 in hippocampal tissue were analyzed by TUNEL staining and western blotting,respectively.SPSS 20.0 software was used for statistical analysis.Results As compared with the propofol group,the rats in the propofol+EMF group had lower escape latency and suimming distance(P<0.05),higher crossing platform times(P<0.05),significantly lower serum Aβand NSE expression levels(P<0.05),statistically higher hippocampal NGF and BDNF expression levels(P<0.05),lower number of apoptotic cells in hippocampus and caspase-3 expression level(P<0.05)and significantly higher Bcl-2 expression levels(P<0.05).There were no statistical differences in escape latency,swimming distance,times of crossing platform and expression levels of Aβ,NSE,NGF,BDNF,Bcl-2 and caspase-3 in the propofol+EMF group rats compared to the control group(P>0.05).Conclusion Low-intensity alternting EMF can significantly improve propofol-induced cognitive function decline in aging rats,which is associated
