摘  要:K_(ATP)通道是一种重要的代谢敏感性离子通道,在生理和病理机制中发挥着重要作用,是一些常见代谢疾病,如糖尿病、心肌缺血、心率失常等的药物作用靶点。该文通过施加特异性开放剂或抑制剂的方法诱导或阻断K_(ATP)通道,采用膜片钳全细胞实验方法检测豚鼠新室肌细胞膜K_(ATP)通道(sarcK_(ATP))的电流密度。mitoK_(ATP)位于胞内线粒体膜上,可通过改变线粒体膜电位进而增强黄素荧光蛋白的自发荧光,利用激光共聚焦显微镜检测黄素荧光蛋白荧光强度的变化来间接测量线粒体K_(ATP)通道的活动。此外,诱导sarcK_(ATP)通道开放可缩短动作电位时程,降低静息膜电位,对心率有重要调节作用。K_(ATP) channel is an important metabolic sensitive ion channel,which plays an important role in physiological and pathological mechanisms.It is often used as a drug target for some metabolic diseases,such as diabetes mellitus,myocardial ischemia,arrhythmia,etc.The sarcK_(ATP)channel is induced by specific openers and blocked by inhibitors,and its current activity is detected directly by whole cell patch clamp technique.The mitoK_(ATP) activity introduced by its opener is identified by examining the spontaneous fluorescence of flavin protein through laser confocal microscope.Besides,the opening of sarcK_(ATP) shortened the action potential duration and decreased the rest membrane potential of guinea pig ventricular myocytes,which plays an important role in regulating heart rate.
