摘 要:该文利用汞特异性核酸适配体和可以嵌入双螺旋脱氧核糖核酸中的荧光染料PicoGreen建立了一种快速检测汞离子的荧光分析方法。通过荧光信号强度来测定汞离子浓度,并获得了汞离子浓度的对数和荧光强度之间的线性关系。方法的线性范围为0.001~100μg/mL,检出限达到0.38 ng/mL。在As^(3+)、Pb^(2+)、Cd^(2+)、Cr^(2+)、K^(+)等干扰离子存在的情况下,具有很好的特异性。同时,在灌溉水实际样品检测中的回收率为95.2%~97.9%,相对标准偏差为2.3%~4.0%。该方法利用核酸适配体和荧光染料实现了对汞离子的痕量检测,是一种简单、快速、灵敏的测定方法。In this paper,a rapid fluorescence assay for the detection of mercury ions was developed using mercury-specific aptamer and PicoGreen,a fluorescent dye embedded in double helix DNA.Mercury ion concentration was determined by fluorescence signal intensity,and the linear relationship between the logarithm of mercury ion concentration and fluorescence intensity was obtained.The linear range of the method was 0.001-100μg/mL,and the detection limit was 0.38 ng/mL.In the presence of interference ions such as As^(3+),Pb^(2+),Cd^(2+),Cr^(2+),K^(+),it has a good specificity.Meanwhile,the irrigation water samples with different concentrations of Hg2+showed a good recovery of 95.2%-97.9%and the relative standard deviation of 2.3%-4.0%.The method is simple,rapid and sensitive,and is applicable for the determination of mercury ions by using aptamer and fluorescent dye.
