摘 要:骨质疏松症是骨代谢紊乱、骨微结构被破坏、骨骼脆性增加,致使容易造成骨折的代谢性骨病综合征。越来越多的证据表明,连接蛋白介导的细胞间隙间通道促进了骨骼生物学的许多方面,包括骨骼发育、骨骼稳态的维持及骨骼细胞对多种细胞外信号的反应性。细胞间隙连接蛋白43(Cx43)是骨骼中存在含量最丰富的间隙连接蛋白,已被证明是骨骼动态平衡不可或缺的组成部分。大量研究表明,细胞间隙连接蛋白43影响成骨细胞和骨细胞的功能,最终影响骨量获得和骨稳态。细胞间隙连接蛋白43发挥功能途径多且复杂,目前关于细胞间隙连接蛋白43调节稳态的分子机制尚不十分清楚,文章对此进行综述。Osteoporosis is a metabolic bone disease syndrome in which bone metabolism is disordered,bone microstructure is destroyed,and bone fragility is increased,which makes it easy to cause fractures.More and more evidences show that connexin-mediated intercellular channels promote many aspects of bone biology,including bone development,the maintenance of bone homeostasis,and the responsiveness of bone cells to a variety of extracellular signals.Connexin 43(Cx43)is the most abundant connexin in bones,and it has been proven to be an indispensable part of bone homeostasis.A large number of studies have shown that connexin 43 affects the functions of osteoblasts and osteocytes,and ultimately affects bone mass acquisition and bone homeostasis.There are many and complex ways of connexin 43 to function.At present,the molecular mechanism of connexin 43 regulating homeostasis is not very clear.This paper reviews the research progress of connexin 43 in osteoporosis.
