一株诱导粘细菌子实体形成的被捕食菌的筛选及分子鉴定

摘  要:为筛选适宜于诱导粘细菌子实体形成的被捕食菌,利用诱导分离的方法,从酵母菌、革兰氏阴性菌及革兰氏阳性菌共30株菌中进行筛选,利用扫描电镜观察形态特征,通过16S rRNA确定系统分类地位,并对其分离培养基进行优化。获得一株被捕食菌GIM1.767,经透射电子显微镜观察长约为1.2—1.24μm,宽度约为0.6—0.65μm,无鞭毛,分子鉴定为Advenella kashmirensis;对分离培养基进行优化,发现添加1%酵母粉的WCX培养基,对粘球菌属能更好的分离诱导。分离得到菌株Advenella kashmirensis,对挖掘粘细菌资源提供一些参考价值。In order to screen the predatory bacteria suitable for inducing the formation of myxobacteria fruiting bodies,the induced separation method was performed to select 30 stains from microbes including yeasts,Gram negative and Gram positive bacteria.Subsequently,the morphological characteristics were observed by scanning electron microscope and phylogentic position was analyzed based on 16S rRNA.In addition,the isolation medium was optimized.One prey bacterium GIM1.767 was identified as Advenella kashmirensis.According to scanning electron microscope,its length and width were 1.2-1.24μm and 0.6-0.65μm,respectively,without flagella.Optimization of isolation medium found that the WCX medium with 1%yeast extract was effective to separate the Myxococcus strains.Isolation of strain A.kashmirensis had certain reference application value to excavate myxobacteria resources..
