摘 要:为探究IGF-1基因和IGF-2基因在关岭牛不同组织中的表达规律,本实验以关岭牛为研究对象,采集关岭牛的肝脏、脾脏、心脏、肺脏、肾脏以及背最长肌6个不同组织,提取总RNA,并采用qRT-PCR技术分析IGF-1基因和IGF-2基因mRNA在关岭牛6个不同组织中的表达水平。结果显示:IGF-1基因在关岭牛6个不同组织中均有表达,与其他5个组织相比,肺脏表达量极显著高于所有组织,IGF-1基因在背最长肌的表达量低于其他5个组织,肝脏的表达量显著高于背最长肌,其余各组织间均无显著差异;IGF-2基因在关岭牛6个不同组织中均有表达,在肺脏中的表达量极显著高于其他5个组织,其次是肝脏,最低为背最长肌,在肝脏组织中的表达量显著高于脾脏和背最长肌,其余组织间无显著差异。本研究揭示了IGF-1基因和IGF-2基因在关岭牛中的表达差异性,为探究关岭牛的生长发育机制提供了基础数据。In order to study the expression levels of IGF-1 and IGF-2 genes in different tissues of Guanling cattle,six different tissues of liver,spleen,heart,lung,kidney and longissimus dorsi were collected from Guanling cattle.The total RNA was extracted and the expression levels of IGF-1 gene and IGF-2 gene mRNA in six different tissues of Guanling cattle were analyzed by qRT-PCR.The results showed that IGF-1 gene was expressed in six different tissues of Guanling cattle.Compared with the other five tissues,the expression of IGF-1 gene in lung was significantly higher than that in all tissues,the expression of IGF-1 gene in longissimus dorsi was lower than that in other five tissues,and the expression in liver was significantly higher than that in longissimus dorsi,but there was no significant difference among other tissues.IGF-2 gene was expressed in six different tissues of Guanling cattle,the expression in lung was significantly higher than that in other five tissues,the second was liver,the lowest was longissimus dorsi,and the expression in liver was significantly higher than that in spleen and longissimus dorsi,but there was no significant difference among other tissues.This study revealed the difference of expression of IGF-1 gene and IGF-2 gene in Guanling cattle,which provided basic data for exploring the growth and development mechanism of Guanling cattle.
