摘  要:目的研究基于"毒瘀交阻"理论组方金果胃康胶囊对胃癌前病变大鼠模型Wnt/Lgr5信号通路调控的分子机制。方法选用SD雄性大鼠随机分为空白组、模型组、金果胃康胶囊(高、中、低剂量)治疗组、维酶素治疗组,我们使用MNNG结合饥饱失常等综合造模法复制PLGC大鼠病理模型,确定模型复制成功,连续灌胃治疗6周后,使用蛋白免疫印迹法检测大鼠胃粘膜组织Wnt/Lgr5信号通路基因及蛋白表达水平。结果与模型组相比较,金果胃康胶囊治疗组大鼠胃黏膜上皮组织中Wnt1mRNA、β-cateninmRNA、Lgr5mRNA表达水平均显著降低,有统计学意义(P<0.01)。结论金果胃康胶囊能够有效治疗PLGC,降低胃癌发生风险,可能与通过调控Wnt/Lgr5信号通路,降低大鼠胃黏膜组织Wnt1、β-catenin、Lgr5的蛋白表达及基因转录相关。Objective To study the molecular mechanism of the regulation of Wnt/Lgr5 signaling pathway in the model of gastric cancer rats based on the theory of“Toxin Stasis Combined Obstruction”.Methods SD male rats were randomly divided into blank group,model group,Jingguo Weikang Capsule(high,medium and low dose)treatment group and vitamin enzyme treatment group.We used MNNG combined with the comprehensive model making method of starvation and satiety to copy the pathological model of PLGC rats,and confirmed that the model was successfully copied and treated continuously for 6 weeks,The Wnt/Lgr5 signaling pathway gene and protein expression level in gastric mucosa of rats were detected by Western blot.Results:Compared with the model group,the expression levels of Wnt1 mRNA,β-catenin mRNA,Lgr5mRNA in the gastric mucosa epithelium of the rats in the treatment group was treated with Jinguo Weikang Capsule were significantly reduced(P<0.01).Conclusion Jinguo Weikang Capsule can effectively treat PLGC and reduce the risk of gastric cancer.It may be related to the reduction of Wnt1,β-catenin mRNA,Lgr5mRNA expression and gene transcription in gastric mucosa of rats by regulating Wnt/Lgr5 signaling pathway.
