摘 要:目的观察载银二氧化钛(Ag-TiO_(2))和Ag-TiO_(2)抗菌涂层气管导管(ETT)对金黄色葡萄球菌生物被膜(BF)的抑制作用。方法采用二甲氧唑黄(XTT)比色法检测Ag-TiO_(2)抗金黄色葡萄球菌BF的最低抑菌浓度(MIC);制备Ag-TiO_(2)涂层ETT,按浓度梯度分为Ag-TiO_(2)11 mg/L组、8 mg/L组、5 mg/L组、2 mg/L组和空白组,分别浸渍于1.0×10^(9) cfu/L浓度的金黄色葡萄球菌菌悬液,通过检测ETT上细菌菌落数及BF的含量,确定抗菌涂层ETT对金黄色葡萄球菌BF形成的影响。结果Ag-TiO_(2)对金黄色葡萄球菌BF具有明显的抑制作用,且呈剂量依赖性,其MIC为10 mg/L;Ag-TiO_(2)涂层ETT具有明显的抗金黄色葡萄球菌BF作用,且浓度越高作用越强,5 mg/L、8 mg/L、11 mg/L组吸光度(A)值均明显低于空白组(0.176±0.004、0.147±0.002、0.094±0.002比0.267±0.045,均P<0.05),Ag-TiO_(2)涂层浓度为2、5、8、11 mg/L的Ag-TiO_(2)涂层ETT对金黄色葡萄球菌的抑制率逐渐升高,且11 mg/L的ETT抗BF效果最好,抑制率分别为6.4%、34.1%、44.9%、64.8%。结论Ag-TiO_(2)和Ag-TiO_(2)涂层ETT对金黄色葡萄球菌BF均有显著的抑制作用。Objective To observe the antibacterial effect of Ag+-loaded TiO_(2)(Ag-TiO_(2))and Ag-TiO_(2) coated endotracheal tube(ETT)on the bacterial biofilm(BF)of Staphylococcus aureus.Methods 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide(XTT)colorimetric method was used to detect minimal inhibitory concertation(MIC)of Ag-TiO_(2) for inhibition of BF of Staphylococcus aureus.The Ag-TiO_(2) coated ETT were prepared,and divided into 11 mg/L,8 mg/L,5 mg/L,2 mg/L and 0 mg/L ETT group,according to the concentration gradient,then impregnated in the liquid with Staphylococcus aureus at a concentration of 1.0×10^(9)cfu/L.The influence of antibacterial coated ETT on the formation of Staphylococcus aureus BF was determined by detecting the colonies of bacteria and BF on the ETT.Results Ag-TiO_(2) had a significant inhibitory effect on Staphylococcus aureus BF in a concentration-dependent manner,and its MIC was 10 mg/L.Ag-TiO_(2) coated ETT has significant anti-Staphylococcus aureus BF effect,and the higher the concentration,the stronger the effect.The absorbance(A)values of Ag-TiO_(2)5 mg/L,8 mg/L,11 mg/L ETT groups were significantly lower than that in control group(0.176±0.004,0.147±0.002,0.094±0.002 vs.0.267±0.045,all P<0.05).The inhibitory rates of Ag-TiO_(2)2 mg/L,5 mg/L,8 mg/L ETT groups were increased gradually,and 11 mg/L Ag-TiO_(2) coated ETT group had the highest inhibitory rate for BF,the inhibitory rates were 6.4%,34.1%,44.9%and 64.8%,respectively.Conclusion Both Ag-TiO_(2) and Ag-TiO_(2) coated ETT have significant inhibitory effects on Staphylococcus aureus BF.
