摘  要:[目的]探讨沉默LncRNA TUG1对骨肉瘤细胞放射敏感性的影响。[方法]采用qRT-PCR检测人成骨细胞hFOB1.19、骨肉瘤细胞HOS、放射抵抗的骨肉瘤细胞HOS-R中TUG1与miR-328-3p的表达量;采用不同方式检测沉默TUG1与miR-328-3p过表达对HOS-R细胞放射敏感性的影响;TUG1的靶基因miR-328-3p;沉默TUG1对HOS-R细胞中miR-328-3p表达量的影响;HOS-R细胞凋亡率;抑制miR-328-3p联合沉默TUG1对HOS-R细胞放射敏感性及细胞凋亡率的影响。[结果]与hFOB 1.19细胞相比,TUG1在HOS与HOS-R细胞中表达量较高(P<0.05),miR-328-3p的表达量显著较低(P<0.05),且在HOS-R细胞中显著低于HOS细胞(P<0.05);沉默TUG1或过表达miR-328-3p显著降低HOS-R细胞存活分数(P<0.05);并增加细胞放射敏感性;TUG1可吸附miR-328-3p并调节其表达水平;抑制miR-328-3p联合沉默TUG1可逆转沉默TUG1对HOS-R细胞放射敏感性的增强作用。[结论]本研究表明LncRNA TUG1可通过靶向调控miR-328-3p的表达促进骨肉瘤细胞凋亡,并抑制其存活,从而增加骨肉瘤细胞的放射敏感性。[Objective] To explore the effect of silencing LncRNA TUG1 on radiosensitivity of osteosarcoma cells. [Methods] The expression levels of TUG1 and miR-328-3 p in human osteoblasts(HFOB 1.19), osteosarcoma cells(HOS) and radio-resistance osteosarcoma cells(HOS-R) were detected by qRT-PCR, while the effect of silenced TUG1, miR-328-3 p overexpression, inhibition of miR-328-3 p combined with silencing of TUG1 on the radiosensitivity and apoptosis of HOS-R cells were detected by different methods. [Results] Compared with HFOB 1.19 cells, the expression of TUG1 was significantly increased in HOS and HOS-R cells(P<0.05), which was significantly higher in HOS-R cells than in HOS cells(P<0.05). In addition, the expression of miR-328-3 p was significantly decreased in HOS cells compared with HFOB 1.19 cells(P<0.05), which was significantly lower in HOS-R cells than in HOS cells(P<0.05). Silencing TUG1 or overexpression of miR-328-3 p significantly reduced the survival score of HOS-R cells, whereas increased the radiosensitivity of the cells.TUG1 adsorbed miR-328-3 p and regulated its expression level. Inhibition of miR-328-3 p combined with the silencing of TUG1 reversed the enhancement of radiosensitivity of silencing TUG1 in HOS-R cells. [Conclusion] LncRNA TUG1 does increase the radiosensitivity of osteosarcoma cells by targeting the expression of miR-328-3 p to promote apoptosis and inhibit cell survival in this study.
