摘  要:目的:建立三叉苦茎的HPLC指纹图谱,为三叉苦药材质量控制提供参考。方法:采用Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm, 5μm),以甲醇-0.2%磷酸溶液为流动相,梯度洗脱;柱温为30℃;检测波长为240 nm;进样量为20μL;采用"中药色谱指纹图谱相似度评价系统(2004 A版)"计算相似度,SPSS 22.0统计软件进行聚类分析和主成分分析,比较三叉苦不同部位(根、茎、带叶嫩枝)的HPLC色谱图。结果:建立了三叉苦茎的HPLC指纹图谱,确定了13个共有峰。28批三叉苦茎指纹图谱与对照图谱的相似度为0.225~0.990,其中22批样品的相似度在0.900以上,28批样品聚为3类。三叉苦茎HPLC色谱图与根相似,与带叶嫩枝差别较大。结论:三叉苦茎HPLC指纹图谱的建立和不同部位色谱图的比较可为三叉苦药材质量评价提供依据。Objective: To establish the HPLC fingerprint of stem from Melicope pteleifolia,so as to provide the basis for its quality control.Methods: The chromatographic separation was performed on a Agilent ZORBAX SB-C18 column(250 mm×4.6 mm, 5 μm),the mobile phase was methanol-0.2% phosphoric acid with gradient elution.The column temperature was 30 ℃,detection wavelength was 240 nm, injection volume was 20 μL.The fingerprint was compared for similarity by using "Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2004 A)",cluster analysis and principal component analysis were adopted in SPSS 22.0,the HPLC chromatograms from different parts of Melicope pteleifolia,including root, stem and twig with leaf were compared.Results: The HPLC fingerprint of Melicope pteleifolia stem was established and 13 common peaks were designated.The similarities of the 28 batches of samples were from 0.225 to 0.990 compared with reference spectrum, among them, 22 batches of samples had a similarity ratio of more than 0.900.The 28 batches of samples were clustered into 3 categories.The HPLC chromatogram of Melicope pteleifolia stem was similar to that of root, but different from that of twig with leaf.Conclusion: The establishment of HPLC fingerprint of Melicope pteleifolia stem and comparison of HPLC chromatogram for different parts can provide the scientific basis for quality evaluation of Melicope pteleifolia.
