摘 要:目的在肝癌H22荷瘤病理状态下观测煎汁煮制炮制雷公藤的减毒增效(或存效)作用及其机制。方法通过对H22荷瘤小鼠抑瘤率、血清生化指标、以及毒性靶器官肝和肾的抗氧化和炎症相关指标的比较分析,评价煎汁煮制炮制雷公藤的减毒增效(或存效)作用及其机制。结果雷公藤生品引起了肝癌H22荷瘤小鼠血清的丙氨酸氨基转移酶、肌酐和尿素氮、毒性靶器官肝和肾的炎性介质肿瘤坏死因子-α等指标的显著升高,以及抗氧化物谷胱甘肽、谷胱甘肽-S-转移酶、谷胱甘肽过氧化物酶、超氧化物歧化酶、过氧化氢酶以及抗炎性介质白介素-10等指标的显著降低;庆幸的是,经金银花、白芍、金钱草、甘草、绿豆分别煎汁煮制炮制后,对雷公藤生品引起的以上指标的异常均有不同程度地逆转作用。此外,各炮制品能使雷公藤对肝癌H22荷瘤小鼠的抑瘤率分别升高21.2%、15.6%、29.9%、18.2%、29.1%。结论煎汁煮制炮制降低了雷公藤的肝毒性和肾毒性,并保存甚或增强了雷公藤的抗肿瘤活性,其中尤以金钱草煎汁煮制炮制的作用为佳,其减毒增效(或存效)的生物机制涉及增强抗氧化防御并抑制炎症反应。Objective To observe the effect and mechanism of detoxification and synergism(or preservation efficacy) of Leigongteng(Tripterygium wilfordii,LGT) prepared by decocting method under hepatoma H22 tumor-bearing pathological state. Method In H22 tumor-bearing mice, through the comparative analysis of tumor inhibition rate, serum biochemical indicators and antioxidant and inflammation-related indicators of toxic target organs(liver and kidney), the attenuating and synergistic effects of decocting method of LGT and its biological mechanism were evaluated. Result In hepatoma H22 tumor-bearing mice, LGT raw products caused significant increase in serum alanine aminotransferase, creatinine and urea nitrogen, tumor necrosis factor-α and other indicators in liver and kidney, as well as glutathione, glutathione glutathione-S-transferase, glutathione peroxidase, superoxide dismutase, catalase and anti-inflammatory mediator interleukin-10 were significantly reduced. Fortunately, through Jinyinhua(Lonicerae Japonicae Flos), Baishao(Paeoniae Radix Alba), Jinqiancao(Lysimachiae Herba), Gancao(Glycyrrhizae Radix et Rhizoma) and Ludou(mung bean) which are separately prepared by the method of decoction cooking, it can significantly reverse the abnormality of the above indicators caused by LGT. In addition, all the processed products can increase the inhibition rate of LGT on hepatoma H22 tumor-bearing mice by 21.2%, 15.6%, 29.9%, 18.2% and 29.1%, respectively. Conclusion Processing by decocting method reduces the hepatotoxicity and nephrotoxicity of LGT, and preserves or even enhances the anti-tumor activity of LGT. Among them, the effect of JQC decocting method is better. The biological mechanism of preservation efficacy involves enhancing antioxidant defense and inhibiting inflammatory response.
