摘  要:目的探讨迷迭香酸(RA)对三聚氰胺(Mel)及其同系物(三聚氰酸混悬液)联合染毒小鼠泌尿生殖系统的影响。方法按照体重将ICR小鼠随机分成3组:正常组、模型组和实验组,每组10只。用隔日染毒造模,用Mel和三聚氰酸混悬液(均为25 mg·kg^(-1)·d^(-1))灌胃,每2天1次;造模的同时,实验组予以RA水溶液(100 mg·kg^(-1)),正常组和模型组予以生理盐水,每天灌胃1次,连续30 d。用比色法检测各组小鼠血清肌酸酐(Cr)、尿素氮(BUN)、尿酸(UA)和丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,用苏木精-伊红(HE)染色观察小鼠肾和睾丸的病理学形态并进行评分,用液相色谱串联质谱法(LC-MS/MS)法测定各组小鼠肾组织中Mel含量。结果正常组、模型组和实验组小鼠的Cr水平分别为(53.71±9.08),(55.82±12.36)和(53.44±7.24)μmol·L^(-1);这3组的BUN水平分别为(6.04±1.31),(6.48±1.57)和(5.87±1.23)mmol·L^(-1);这3组的UA水平分别为(199.50±20.54),(205.17±76.34)和(202.93±11.75)μmol·L^(-1);这3组的MDA水平分别为(9.99±1.12),(10.25±1.37)和(9.26±1.09)mmol·mL^(-1);这3组的SOD水平分别为(162.43±7.41),(172.03±18.14)和(171.39±6.97)U·mL^(-1),实验组与模型组比较,上述指标的差异均无统计学意义(均P>0.05)。正常组、模型组和实验组的肾结石评分分别为0,(2.30±0.82)和(0.78±0.67)分;这3组的肾小管损伤评分分别为(0.42±0.07),(2.70±0.48)和(1.11±0.61)分,实验组与模型组比较,上述指标的差异均有统计学意义(均P<0.05)。这3组的小鼠肾Mel含量分别为0,6.9,5.7 ng·g^(-1),实验组低于模型组。结论RA可减少Mel在体内的蓄积,抑制肾结石的形成,并能减轻对肾小管和睾丸的损伤。Objective To explore the protective effect of rosmarinic acid(RA)on genitourinary system of mice induced by melamine and its homologues.Methods The ICR mice were randomly divided into three groups according to weight:normal group,model group and experimental group,with 10 mice in each group.The model group and experimental group were given melamine and cyanuric acid suspension(both 25mg·kg^(-1)·d^(-1))by intragastric administration to establish the kidney stone model,once every two days,while the normal group was given the same volume of normal saline.During the process of establishing the mice model,the experimental group was given RA(100 mg·kg^(-1))every afternoon,while the normal group and model group were given normal saline.All mice were administered orally for 30 d.The contents of the creatinine(Cr),urea nitrogen(BUN),uric acid(UA),malondialdehyde(MDA)and superoxide dismutase(SOD)activity were measured.Pathological morphology of renal and testicular tissue were observed by hematoxylin-eosin(HE)and score.The contents of melamine in kidney were determined by liquid chromatography-tandem mass spectrometry(LC-MS/MS).ResultsThe serum Cr in normal group,model group and experimental group were(53.71±9.08),(55.82±12.36),(53.44±7.24)μmol·L^(-1),respectively;BUN in the 3 groups were(6.04±1.31),(6.48±1.57),(5.87±1.23)mmol·L^(-1),respectively;UA in the 3 groups were(199.50±20.54),(205.17±76.34),(202.93±11.75)μmol·L^(-1),respectively;the MDA in the 3 groups were(9.99±1.12),(10.25±1.37),(9.26±1.09)mmol·L^(-1),respectively;SOD in the 3 groups were(162.43±7.41),(172.03±18.14),(171.39±6.97)U·m L^(-1);compared between experimental group and model group,the difference of the above indexes were not significant(all P<0.05).The renal crystal deposition scores in normal group,model group and experimental group were 0,(2.30±0.82),(0.78±0.67)point,respectively;the score of renal tubular injury in the 3 groups were(0.42±0.07),(2.70±0.48)and(1.11±0.61)point,respectively.Compared between model group and n
