摘 要:为探讨过敏性鼻炎(allergic rhinitis,AR)对机体Th9变化及相关细胞因子表达的影响,收集2018年1月至12月于南宁市第二人民医院就诊的AR患者46例为病例组,同期健康体检者12例为健康组,采集两组研究对象血清,检测IL-9、IL-4和TGF-β1表达水平。取C57BL/6小鼠6只,采用随机数字表法分为模型组和对照组(n=3),建立卵清蛋白(ovalbumin,OVA)致敏的AR小鼠模型,根据鼻部症状学评分判定模型构建是否成功。qPCR检测各组小鼠鼻黏膜组织中PU.1、IRF-4、IL-9、IL-4和TGF-β1转录水平;FACS检测小鼠外周血Th9百分比。结果显示,与健康组比较,病例组血清样本中IL-9、IL-4和TGF-β1的表达水平显著升高(均P<0.01);在小鼠模型中,与对照组相比,模型组小鼠鼻黏膜组织中PU.1、IRF-4、IL-9、IL-4和TGF-β1 mRNA表达水平显著升高(均P<0.05),Th9百分比也显著升高(P<0.05)。由此,Th9及相关炎性因子在AR疾病发生发展过程中可能发挥一定作用。In order to explore the relationship between allergic rhinitis(AR)and Th9percentage,cell cytokines expressions,46AR patients from Nanning Second People's Hospital and 12healthy controls were collected from January to December,2018.Serum was collected to detect IL-9,IL-4and TGF-β1expression levels.Six C57BL/6mice were divided into the model group and control group by random number table(n=3,respectively).The AR mouse model sensitized by ovalbumin(OVA)was established and confirmed according to the nasal symptom score.qPCR was used to detect the mRNA expression levels of PU.1,IRF-4,IL-9,IL-4 and TGF-β1 in each group mice nasal mucosa.The Th9percentage in peripheral blood was measured by FACS.The results showed that the expression levels of IL-9,IL-4and TGF-β1were significantly increased in the patient group than those in the healthy group(all P<0.01);In the animal experiment,compared with the control group,the mRNA expression levels of PU.1,IRF-4,IL-9,IL-4 and TGF-β1in the nasal mucosa of the model group were significantly increased(all P<0.05),and the Th9percentage was also significantly increased(P<0.05).Therefore,Th9and related inflammatory factors may play an important role in the occurrence and development of AR disease.
