摘  要:目的探讨水杨梅黄酮类成分的纯化工艺、表征及抗氧化作用。方法以水杨梅总黄酮为指标性成分,比较大孔吸附树脂法、石灰乳-硫酸法及萃取法对水杨梅总黄酮的分离纯化效果,筛选最佳纯化工艺。采用超高效液相色谱-四极杆飞行时间串联质谱(UPLCQ-TOF/MS)法检测纯化前后的水杨梅黄酮类成分,对其进行初步表征。采用铁还原法、ABTS自由基清除率(以维生素C为阳性对照)、DPPH自由基清除率考察水杨梅总黄酮的抗氧化活性。结果水杨梅最佳纯化工艺为以HPD-100型大孔树脂吸附,50%乙醇为洗脱剂;纯化前、后分别检测出789个和730个化学成分,表征后分别得到36个和28个黄酮类成分;铁还原法测得样品溶液吸光度为0.7433(阳性对照为0.8233),ABTS自由基清除率为84.89%,DPPH自由基清除率为48.69%。结论水杨梅总黄酮可采用大孔吸附树脂法纯化,操作简单,检测有效,且水杨梅有一定的抗氧化作用。Objective To investigate the purification process,characterization and antioxidant activity of flavonoids of Adina rubella.Methods The effect of the macroporous adsorption resin method,lime milk-sulfuric acid method and extraction method on the separation and purification of total flavones of Adina rubella were compared with total flavones of Adina rubella was taken as the index component,and the optimum purification process was screened.The flavonoids of Adina rubella before and after purification were detected and Preliminarily characterized by the ultra high-performance liquid chromatography-time of flight mass spectrometry(UPLC-Q-TOF/MS).The antioxidant activity of total flavones of Adina rubella was investigated by iron reduction method,ABTS free radical scavenging rate(vitamin C was taken as the positive control)and DPPH free radical scavenging rate.Results The optimum purification process of Adina rubella was as follows:HPD-100 macroporous resin was used for adsorption with 50%ethanol as eluent.A total of789 and 730 chemical components were detected before and after purification,36 and 28 flavonoids were obtained after characterization.The absorbance of the sample solution measured by the iron reduction method was 0.7433(that of positive control was 0.8233),the free radical scavenging rate of ABTS was 84.89%,and the free radical scavenging rate of DPPH was 48.69%.Conclusion The total flavones of Adina rubella can be purified by the macroporous adsorption resin method,which is simple and effective,and Adina rubella has a certain antioxidant activity.
