摘 要:为了解福建省猪圆环病毒4型(PCV4)流行株的现状和遗传演化,收集了2018年-2019年福建省规模化猪场200份疑似断奶仔猪多系统衰竭综合征(PMWS)的猪组织病料或血清,通过荧光定量PCR方法进行PCV4的核酸检测。结果表明,200份检测样品中PCV4核酸阳性的有4份,检出率为2%(4/200),成功克隆了1个PCV4全基因组序列(FJ-PCV4)。FJ-PCV4的基因组全长为1 770 bp,与国内PCV4代表毒株(GenBank登录号MK948416和MK986820)核苷酸序列同源性为98.8%~99.2%,与PCV1、PCV2及PCV3代表毒株Eng-1970、AF055392、AF055394、DK1980PMWSfree、BDH、 MN2016和MO2015等的核苷酸同源性为43.3%~52.0%,而与水貂圆环病毒Mink circovirus strain SD16、Mink circovirus strain MiCV-DL13同源性为68.1%~68.2%,表明PCV4是一种不同于PCV1、PCV2和PCV3的新型圆环病毒。研究结果为PCV4的分子流行病学研究提供了基础资料。In order to understand the current status and genetic evolution of porcine circovirus type 4 strains in Fujian province, a total of 200 samples(kidneys, lungs, lymph nodes and blood) with symptoms of postweaning multisystemic wasting syndrome(PMWS) collected from farms of Fujian province from 2018 to 2019 and these samples were used to detect PCV4 by real-time PCR.The results showed that the infection rate of PCV4 in pig farms was only 2%(4/200),and the full-length genome of FJ-PCV4 isolate was successfully amplified.The full-length genome of FJ-PCV4 is 1 770 bp, which shared 98.8%-99.2% nucleotide identity with Chinese reference PCV4 isolates(GenBank No.MK948416 and MK986820),but only 43.3%-52.0% with PCV1,PCV2 and PCV3 reference isolates(Eng-1970,AF055392,AF055394,DK1980 PMWSfree, BDH,MN2016 and MO2015),in contrast, 68.1%-68.2% with mink circovirus(Mink circovirus strain SD16 and Mink circovirus strain MiCV-DL13),indicating that PCV4 is a novel circovirus different from PCV1,PCV2 and PCV3.Our data will help in understanding the evolutionary characteristics of PCV4 and effective prevention and control of PCV4.
